Fas/FasL系统在子宫内膜异位症中的表达与意义 被引量:5

摘要 目的 探讨Fas和Fas配体 (FasL)在子宫内膜异位症发病中的作用。方法 采用免疫组化链霉菌抗生物素蛋白 -过氧化物酶染色法 (S -P法 )检测子宫内膜异位症 5 6例的异位内膜与在位内膜 (30例 )中Fas和FasL蛋白的表达 ;采用原位杂交方法检测FasLmRNA水平。设正常子宫内膜为对照组 (2 5例 )。结果 Fas蛋白在卵巢异位症 (OEM )与子宫腺肌症 (AM )组的阳性表达率分别为 6 3%和 77% ,均低于正常对照组 (92 % ) ,差异有显著性 (P <0 .0 5 )。FasL蛋白在OEM与AM组中的阳性表达率分别为 97%和 73% ,明显高于对照组 (32 % ) ,差异有极显著性 (P <0 .0 1) ,且二者的表达均不随月经周期而改变。FasLmRNA的表达强度在异位症组 (包括OEM和AM两组 )亦明显高于对照组 ,差异有显著性 (P <0 .0 1,P <0 .0 5 )。结论 Fas/FasL系统中FasL基因转录激活 ,使Fas与FasL蛋白表达比例失衡 ,可能对异位内膜组织逃避免疫监视系统具有重要作用。 Objective To explore the roles of Fas and Fas Ligand(FasL) in the pathogenesis of endometriosis (EM). Methods Immunohistochemical S-P method was employed to detect the expression of Fas and Fas Ligand in ectopic tissues and eutopic endometrium of the patients with EM ( n =56) and the normal controls( n =20). In situ hybridization technique was used to investigate the transcription level and condition of FasL gene. Results The expression level of Fas in ectopic and eutopic endometrium in EM was lower than that in control group( P <0.05), butthe expression level of FasL was higher than that of the control group( P <0.01). And their expressions were irrespective of the phases of menstrual cycle. Results of in situ hybridization showed that the expression level and the positive rate of FasLmRNA in endometriotic tissue were both higher than that in normal endometrium( P <0 01, P <0.05). Conclusion Both Fas and Fas Ligand(FasL) are abnormally expressed in endometriosis and adenomyosis, which may play an important role in the pathogenesis of the diseases.
