系统性红斑狼疮患者血和尿DNaseⅠ与肌动蛋白的相关性研究 被引量:1

Study on the Correlation Between DNaseⅠand G-actin of the Blood and Urine in SLE Patients

摘要 目的探讨系统性红斑狼疮(SLE)患者血和尿中DNaseⅠ活性与肌动蛋白浓度的相关性。方法应用DNA-甲基绿比色法和ELISA间接竞争法检测65例SLE患者和30例正常人对照血、尿中的DNaseⅠ活性和肌动蛋白浓度,分析其相关关系。结果SLE组患者血清、尿DNaseⅠ活性均低于对照组(P<0.01),病情活动期组两者亦均低于缓解期组(P<0.01);肾损害组血清DNaseⅠ活性与非肾损害组比较,差异无显著性,而尿DNaseⅠ活性却降低(P<0.01);SLE组血浆肌动蛋白浓度高于对照组(P<0.05),但尿肌动蛋白浓度两组间差异无显著性;活动期患者血浆肌动蛋白浓度高于缓解期(P<0.01),但尿肌动蛋白浓度两组间差异无显著性;肾损害组血浆肌动蛋白浓度与非肾损害组比较,差异无显著性;但前者尿肌动蛋白浓度却高于后者(P<0.05)。SLE患者血浆肌动蛋白浓度与DNaseⅠ活性之间呈负相关(r=-0.35,P<0.05),而尿肌动蛋白浓度与DNaseⅠ活性之间无相关性(r =-0.23,P>0.05)。结论SLE患者血中DNaseⅠ活性与肌动蛋白浓度之间呈负相关,提示血中肌动蛋白浓度的增高可能是DNaseⅠ活性降低的原因之一。 Objective To measure the DNaseⅠactivity and G-actin concentration of the blood and urine in SLE patients and analyze their correlation.Methods The DNaseⅠactivity and G-actin concentra-tion were measured by DNA-methyl green substrate colorimetry and indirect competitive ELISA in the blood and urine of65SLE patients and30normal controls.Results The DNaseⅠactivity was lower in the sera and urine in SLE patients than that in healthy controls(P<0.01),and also lower in active patients than that in remitted patients(P<0.01).No significant difference was detected in the serum DNaseⅠactivities between the renal involved and non-renal involved patients(P>0.05),however,the urine DNaseⅠactivity was significantly lower in the renal-involved group(P <0.01).The plasma G-actin concentrations were high-er in SLE patients than those in healthy controls(P<0.05),but no significant difference was found in the urine G-actin concentrations between two groups(P >0.05).Active patients had higher plasma G-actin concentrations(P<0.01),but there was no difference in the urine G-actin concentrations(P>0.05)in comparison with patients in remission.No significant difference was found in the plasma G-actin concentra-tions between renal involved and non-renal involved patients(P>0.05);however,the urine G-actin con-centrations were significantly higher in renal involved patients than those of non-renal involved patients(P<0.05).There were negative correlation between the DNaseⅠactivity and G-actin concentration in the blood(r =-0.353,P<0.05),and no correlation between DnaseⅠand G-actin in the urine(r =-0.234,P>0.05)in SLE patients.Conclusions The elevated G-actin concentration in the blood of SLE may inhibits the DNaseⅠactivity,which may be one of possible causes to render the decreased DNaseⅠactivity in the blood.
