摘要
目的 本实验设计合成靶向NF κB的哑铃形圈套ODNs,并验证其抗TNFα生成效应。方法 以阳离子脂质体为载体 ,将圈套ODNs转染大鼠pM细胞 6小时后用LPS(脂多糖 ) 10 μg ml刺激 ,用ELISA法及RT PCR法测圈套ODNs对TNFα蛋白及mRNA水平的影响。结果 哑铃形圈套ODNs可明显降低TNFα在mRNA及蛋白水平的表达。结论 靶向NF κB的哑铃形圈套ODNs具有下调炎性介质TNFα的表达。
Objective To synthetize Decoy ODNs in dumbbell shape with the nuclear sequence similar to NF κB cis elements,and to measure the anti inflammatory action of decoy ODNs targeting NF κB.?Methods With carriers of cationic liposomes,decoy ODNs were transfected into pM cells from rats. Then the inhibitory effects of the decoy ODNs of TNFα were analyzed.?Results Decoy ODNs could decrease the expression of TNFα in a dose dependent manner.?Conclusion Decoy ODNs targeting nuclear factor κB can decrease the expression of inflammatory mediators in pMφ cells.
