摘要
目的 :探讨加温联合钙剂对诱导SK H SN细胞凋亡的作用及其意义。方法 :采用 3种温度和4种浓度的外源性氯化钙 (CaCl2 )诱导SK H SN细胞凋亡。采用形态学观察、原位缺口末端标记(TUNEL)的方法检测细胞凋亡的结果 ,并用火焰原子吸收分光光度法测定细胞内钙含量 ( [Ca]i)。结果 :加温和 /或加钙均可诱导SK H SN细胞凋亡。在有外源性钙剂存在时 ,加温可明显升高 [Ca]i。同一温度下 ,各组的凋亡率 (APO % )和 [Ca]i均随外源性CaCl2 浓度的升高而升高 ;同一CaCl2 浓度下 ,APO %和 [Ca]i均随温度的升高而升高。析因设计的方差分析结果显示 ,加温和加钙在诱导细胞凋亡时有很强的协同作用 ,P <0 0 1。APO %和 [Ca]i之间呈正相关 ,r =0 983,P <0 0 1。结论 :加温和加钙在诱导SK H SN细胞凋亡方面有很强的协同作用 ,APO %和 [Ca]i呈正相关 ,通过增加 [Ca2
Objective To observe the effect of heat and calcium on inducing apoptosis of SK-H-SN cells and it significance.Methods SK-H-SN cells were exposed to three kinds of heat or/and four kinds of calcium.Morphological study and TUNEL method were used to observe apoptosis cells and the APO% was determined.While the i was determined by method of flame atom absorption.Results Hyperthermia or/and calcium can induce apoptosis of SK-H-SN cells.Heat may promote i of SK-H-SN cells with exogenous calcium.In the same temperature,APO% and i was increased when the concentration of CaCl 2 was promoted.And in the same concentration of CaCl 2,the APO% of which was increased when temperature was risen.The result of factorial experiment design shown that heat and calcium has coordinated interaction on inducing apoptosis of SK-H-SN cells,P<0.01,and APO% and i is correlativity,r=0.983,P<0.01.Conclusions Heat and calcium has coordinated interaction on inducing apoptosis of SK-H-SN cells,APO% and i is correlativity.Increasing i may be one mechanism of hyperthermia to induce apoptosis.
