摘要
目的 研究自噬特异性抑制剂 3 甲基腺嘌呤 (3 methyladenine ,3MA)对长春新碱 (vincristine,VCR)诱导的肝癌细胞系HepG2细胞自噬性凋亡的影响。 方法 利用已建立的VCR诱导HepG2细胞自噬性凋亡之模型 ,采用monodansylcadaverin(MDC)染色及流式细胞术荧光强度测定方法对自噬进行定量研究 ,采用流式细胞术及DNA电泳检测细胞凋亡。结果 3MA可以特异性地阻止自噬泡形成 ,还可以部分地抑制HepG2细胞自噬性凋亡。 结论 自噬是VCR诱导的HepG2细胞自噬性凋亡所必需的。
Objective To study the influence of 3-methyladenine(3MA),a special inhibitor of autophagy, on the autophagic apoptosis induced by vincristine (VCR) in cell line HepG2. Methods The autophagic apoptosis induced by VCR in HepG2 cells was used as in vitro models. With measure of monodansylcadaverin(MDC) staining, the autophagy was analyzed quantitatively. Apoptosis was investigated by flow cytometry and electrophoresis of DNA. Results 3MA can specifically inhibit the formation of autophagic vacuoles and can partially prevent the autophagic apoptosis of HepG2 cells. Conclusion Autophagy is needed in HepG2 cells autophagic apoptosis that was induced by VCR.
