摘要
目的探讨抗苗勒氏管激素Ⅱ型受体(AMHR2)在人子宫内膜中的表达情况及其与子宫内膜容受性的相关性。方法选取2016年7-10月在吉林省人民医院生殖医学中心行IVF-ET助孕的患者42例为研究对象,分为正常对照组(22例)和薄型子宫内膜组(20例)。检测患者子宫内膜厚度及雌激素水平,初步评价子宫内膜容受性。在月经后第5~7天进行内膜搔刮,留取内膜组织,提取子宫内膜组织总RNA,荧光定量PCR检测AMHR2、VEGF基因表达水平。采用h CG (100 ng/ml)处理人子宫内膜细胞株ishakawa细胞24 h,免疫印迹法检测AMHR2表达水平。结果薄型子宫内膜组患者子宫内膜组织中AMHR2和VEGF水平均低于正常对照组,分别下降至正常水平的64%和55%,差异有统计学意义(P<0. 05)。h CG处理24h后AMHR2蛋白水平显著增加,差异有统计学意义(P<0. 05)。结论 AMHR2和VEGF在薄型子宫内膜患者的子宫内膜中表达较低,h CG处理能够提高子宫内膜细胞中AMHR2的表达,其水平与子宫内膜容受性具有一定的相关性。
Objective To explore the expression of anti-Müllerian hormone type Ⅱ receptor( AMHR2) in human endometrium and the correlation with human endometrial receptivity. Methods Forty-two patients undergoing IVF-ET in Center for Reproductive Medicine,Jilin Province People’s Hospital from July to October in 2016 were selected and divided into normal control group( 22 patients) and thin endometrium group( 20 patients). Endometrial thickness and estrogen level were detected,endometrial receptivity was preliminarily evaluated. Endometrium scratching was performed from the fifth day to the seventh day after menstruation. Endometrial tissue was obtained to extract total RNA. Fluorescent quantitative PCR was used to detect the expression levels of AMHR2 and VEGF. h CG( 100 ng/ml) was used to treat human endometrial cell line Ishakawa cells for 24 hours. Western blotting was used to detect the expression level of AMHR2.Results The levels of AMHR2 and VEGF in endometrium of thin endometrium group were statistically significantly lower than those in normal control group,which decreased to 64% and 55% of the normal levels,respectively( P<0. 05). The level of AMHR2 protein after h CG treatment for 24 hours increased significantly( P<0. 05). Conclusion The expression levels of AMHR2 and VEGF are low in thin endometrium. h CG treatment can increase AMHR2 expression in endometrial cells,and the level is correlated with endometrial receptivity to a certain extent.
