子痫前期孕妇血清和胎盘中miR-206和FSTL1表达水平及意义 被引量:1

Expression levels and significance of miR-206 and FSTL1 in serum and placenta of pregnant women with preeclampsia

摘要 目的分析子痫前期孕妇血清和胎盘中miR-206、卵泡抑素样蛋白1 (FSTL1)的表达情况并探讨其意义,为评估子痫前期病情的生物学指标提供依据。方法选取2015年3月-2017年2月在青岛市第八人民医院产科行剖宫产术的51例子痫前期孕妇为研究对象,同期选取25例行剖宫产术的正常孕妇作为对照组。采用实时荧光PCR (qRT-PCR)法检测血清和胎盘miR-206水平,采用酶联免疫吸附(ELISA)法检测血清FSTL1蛋白水平,采用免疫印迹(WB)法检测胎盘FSTL1水平;分析miR-206与FSTL1表达的相关性。结果子痫前期孕妇的TG、TC、LDL-C、Apo B及24 h尿蛋白总量均高于对照组孕妇,HDL-C和Apo A低于对照组孕妇,差异有统计学意义(P<0. 05);重度组孕妇的TG、TC、LDL-C、Apo B及24 h尿蛋白总量显著高于轻度组孕妇,HDL-C和Apo A低于轻度组孕妇,差异有统计学意义(P<0. 05)。与对照组孕妇比较,子痫前期孕妇血清及胎盘中miR-206水平上调,FSTL1蛋白水平下调,差异有统计学意义(P<0. 05)。重度组孕妇血清及胎盘中miR-206水平高于轻度组孕妇,FSTL1水平低于轻度组孕妇,差异有统计学意义(P<0. 05)。miR-206表达与子痫前期患者的TG、TC、LDL-C、Apo B及24 h尿蛋白总量呈正相关,与HDL-C、Apo A水平负相关(P<0. 05); FSTL1表达与子痫前期患者的TG、TC、LDL-C、Apo B及24 h尿蛋白总量呈负相关(P<0. 05),与HDL-C、Apo A正相关(P <0. 05)。子痫前期孕妇血清和胎盘中miR-206、FSTL1表达均呈负相关(r=-0. 698,P<0. 05; r=-0. 774,P<0. 05)。结论 miR-206在子痫前期孕妇血清和胎盘中明显高表达,FSTL1在子痫前期孕妇血清和胎盘中明显低表达,二者与子痫前期的发生和严重程度密切相关。 Objective To analyze the expression levels of miR-206 and FSTL1 in serum and placenta of pregnant women with preeclampsia,explore the significance,and provide a basis for evaluating biological indicators of preeclampsia. Methods Fifty-one pregnant women with preeclampsia undergoing cesarean section in the Eighth People’s Hospital of Qingdao from March 2015 to February 2017 were selected as study object,and 25 normal pregnant women undergoing cesarean section in the hospital during the same period were selected as control group. Real-time fluorescence PCR( qRT-PCR) was used to detect the levels of miR-206 in serum and placenta. ELISA was used to detect the levels of serum FSTL1 protein. Western blotting was used to detect the levels of FSTL1 in placenta. The correlation between miR-206 expression and FSTL1 expression was analyzed. Results The levels of TG,TC,LDL-C,Apo B,and 24-hour urine protein in pregnant women with preeclampsia were statistically significantly higher than those in control group,while the levels of HDL-C and Apo A in pregnant women with preeclampsia were statistically significantly lower than those in control group( P<0. 05). The levels of TG,TC,LDLC,Apo B,and 24-hour urine protein in pregnant women with severe preeclampsia were statistically significantly higher than those in pregnant women with mild preeclampsia,while the levels of HDL-C and Apo A in pregnant women with severe preeclampsia were statistically significantly lower than those in pregnant women with mild preeclampsia( P<0. 05). Compared with the pregnant women in control group,the expression levels of miR-206 in serum and placenta of pregnant women with preeclampsia increased,while the expression level of FSTL1 protein decreased,there were statistically significant differences( P< 0. 05). The expression levels of miR-206 in serum and placenta of pregnant women with severe preeclampsia were statistically significantly higher than those in pregnant women with mild preeclampsia,while the expression levels of FSTL1 in serum and placenta of pregnant women with severe preeclampsia were statistically significantly lower than those in pregnant women with mild preeclampsia( P<0. 05). miR-206 expression was positively correlated with TG,TC,LDL-C,Apo B,and 24-hour urine protein( P<0. 05),miR-206 expression was negatively correlated with HDL-C and Apo A( P<0. 05). FSTL1 expression was negatively correlated with TG,TC,LDL-C,Apo B,and 24-hour urine protein( P<0. 05),FSTL1 expression was positively correlated with HDL-C and Apo A( P<0. 05). miR-206 expression was negatively correlated with FSTL1 expression in serum and placenta of pregnant women with preeclampsia( r =-0. 698,P<0. 05;r =-0. 774,P < 0. 05). Conclusion miR-206 is highly expressed in serum and placenta of pregnant women with preeclampsia significantly,and FSTL1 is lowly expressed in serum and placenta of preeclampsia women significantly. miR-206 and FSTL1 are closely related to occurrence and severity of preeclampsia.
