CD4^+ CD25^+ Foxp3^+调节性T细胞在儿童哮喘中的作用研究 被引量:7

摘要 目的探讨CD4^+CD25^+Foxp3^+调节性T细胞(Treg)在儿童支气管哮喘发病中的作用。方法选取2015年8月-2017年7月在该院儿科诊断为哮喘的患儿80例,分为急性发作期(急性组)、缓解期(缓解组)的哮喘儿童,各40例,另选取年龄、性别匹配的健康儿童(对照组) 40例,应用流式细胞术检测CD4^+CD25^+Foxp3^+Treg占CD4^+T百分比,实时荧光定量PCR检测功能基因Foxp3相对表达量。结果急性组CD4^+CD25^+Foxp3^+Treg占CD4^+T百分比(0. 98±0. 92)和Foxp3基因相对表达量(2. 55±1. 37)最低,其次为缓解组(3. 46±1. 53,6. 82±2. 66)和对照组(5. 30±2. 56,15. 33±6. 81),3组比较差异均有统计学意义(均P<0. 05)。结论 CD4^+CD25^+Foxp3^+Treg降低是儿童哮喘的发病机制之一。 Objective To explore the role of CD4^+CD25^+Foxp3^+regulatory T( Treg) cells in onset of asthma in children.Methods Eighty children diagnosed as asthma in Department of Pediatrics in Rizhao Municipal Maternal and Child Health Care Hospital from August 2015 to July 2017 were selected and divided into acute group and remission group,40 children in each group. Forty age-and gender-matched healthy children( control group) were selected as control group. Flow cytometry was used to detect the percentages of CD4^+CD25^+Foxp3^+Treg cells in CD4^+T cells. Real-time fluorescence quantitative PCR was used to detect the relative expression level of functional gene Foxp3. Results In acute group,the percentage of CD4^+CD25^+Foxp3^+Treg cells in CD4^+T cells and the relative expression level of Foxp3 gene were the lowest,which were( 0. 98 ± 0. 92) and( 2. 55 ± 1. 37),respectively,followed by remission group( 3. 46 ± 1. 53,6. 82±2. 66) and control group( 5. 30±2. 56,15. 33± 6. 81),there were statistically significant differences among the three groups( P<0. 05). Conclusion The decrease of CD4^+CD25^+Foxp3^+Treg cells is one of the pathogenesis of pediatric asthma.
