摘要
建立测定维生素K_2软胶囊中的维生素K_2(35)含量的正相高效液相色谱方法。色谱柱为AgilentZorbax CN(250×4.6mm,5μm),流动相为正己烷(0.08%异丙醇),流速为0.5mL/min,检测波长为254nm。样品经异丙醇超声溶解提取后,直接进样测定,外标法定量。结果表明维生素K_2(35)在1.0~20.0mg/L范围内线性关系良好,相关系数r^2=1.000;该方法操作简便、高效快速、回收率高、专属性好,可用于大批量测定维生素K_2软胶囊中维生素K_2(35)的含量。
To establish a method for the determination of Vitamin K2(35) in the capsule by normal-phase high performance liquid chromatography(NP-HPLC), the samples are performed on Agilent Zorbax CN(250×4.6 mm, 5μm) column with 100% hexane and 0.08% propane as mobile phase, the flow rate at 0.5 mL/min, the detection wavelength at 254 nm, and the column temperature at 25℃. The samples are dissolved by ultrasonic with propane, and directly injected into high performance liquid chromatography, quantitative by external standard method. Vitamin K2(35) has a good linearity in the range of 1.0~20.0 mg/L, and the correlation coefficient is 1.000. The method has a good accuracy, good reproducibility and simple operation, and can be applied for the determination of Vitamin K2(35) in the capsule.
