摘要
目的了解湖北省随州市脑膜炎奈瑟菌(Neisseria meningitidis,Nm)的菌群和分子分型特征。方法通过2010-2018年随州市健康人群带菌率调查获得Nm菌株,分别采用玻片凝集法和普通PCR进行血清分群和基因分群;选取部分菌株进行脉冲场凝胶电泳(Pulsed-field gel electrophoresis,PFGE)分型和多位点序列分型(Multilocus sequence typing,MLST)分析。结果 2010-2018年随州市Nm带菌率为0.54%-9.88%,共获得108株Nm菌株。血清分群显示,在所有Nm菌株中B群40株、C群4株、X群1株、血清不可分群63株;基因分群显示,B群64株、C群5株、X群1株、W群1株、cnl菌株6株、基因不可分群(Non-genogroupable,NG)31株。MLST分型显示,31株Nm菌株共有20种ST型,其中14种(25株)可归入克隆群CC4821(17株)、CC198(5株)、CC32(2株)和CC41/44(1株),6种(6株)不能归入现有克隆群。PFGE分型显示,86株Nm菌株可分为47种PFGE带型,其中2018年B群和NG菌株为优势带型,其他菌株呈高度多态性。结论 2010-2018年随州市Nm带菌率呈上升态势,以B群为主要菌群、CC4821为优势克隆群。
Objective To determine groups and molecular types of Neisseria meningitidis(Nm)strains isolated in Suizhou city of Hubei province.Methods We obtained Nm strains in a survey of Nm carriage among a healthy population of Suizhou in 2010-2018 to determine Nm serogroups and genogroups through the slide agglutination test and PCR.We chose Nm strains for analyses of multilocus sequence typing(MLST)and pulsed field gel electrophoresis(PFGE).Results Nm carriage rates were 0.54%-9.88%in Suizhou during 2010-2018.A total of 108 Nm strains were obtained,which included 40,4,1,and 63 strains of B,C,X,and non-serogroupable serogroups,respectively,and 64,5,1,1,6,and 31 strains of B,C,X,W,cnl,and non-genogroupable(NG)genogroups,respectively,MLST analysis of31 Nm strains identified 20 sequence types(STs),among which 14 types(25 strains)were clonal complex CC4821(17 strains),CC198(5 strains),CC32(2 strains),and CC41/44(1 strain);6 types(6 strains)were unassigned complexes.PFGE analysis of 86 Nm strains showed that 47 PFGE types were identified,among which B and NG strains were predominant band types in 2018 and other strains were highly polymorphic.Conclusions The carriage rate of Nm showed an increasing trend in Suizhou during 2010-2018,with B and CC4821 being predominant group and clonal complexes,respectively.
