抗火鸡疱疹病毒特异性血清的制备与检测

Preparation and Identification of Specific Antiserum Against Turkey Herpes Virus for Vaccine Test

抗火鸡疱疹病毒(Turkey herpes virus,HVT)特异性标准血清是鸡HVT活疫苗成品和毒种鉴别检测以及外源病毒检测中常用的标准品。为了制备检测用抗鸡HVT特异性血清,研究选取200只2周龄SPF鸡进行了针对HVT的基础免疫和加强免疫,于第四次免疫后21d采血分离血清,血清经混合、分装、冷冻真空干燥后,按照《中国兽药典》(2015年版)对其进行了性状、细菌、支原体、外源病毒、效价、特异性、剩余水分和真空度检测。结果表明,制备的抗HVT血清特异性良好,其中和效价为1:8,其他各项检测均符合《中国兽药典》(2015年版)规定,可应用于鸡HVT活疫苗成品或毒种的外源病毒检测以及鉴别检测,制备和检验流程对其他禽源病毒阳性血清的制备也具有一定的参考价值。

The specific antiserum against turkey herpes virus(HVT) is commonly used for identification tests and exogenous virus tests of HVT live vaccines or it′s seeds. To prepare the specific antiserum against HVT, 200 SPF chickens of 2 weeks of age were inoculated with HVT. 21 days after the last immunity, serums were collected and mixed thoroughly.After lyophilization, the serum were submitted to character, bacteria, Mycoplasma, exogenous virus, neutralization titer, specificity, residual moisture and vacuum level according to China Veterinary Pharmacopoeia(2015) and all results showed the quality of obtained serum meeted the standards of China Veterinary Pharmacopoeia(2015). The neutralization titer was 1:8;There were no antibodies against other avian disease viruses in this serum using APG, ELISA and HI test.Overall, the antiserum with high specificity and neutralization titer laid a solid foundation for the identification tests and exogenous virus tests of HVT live vaccines, and the process of preparation and testing also provided certain reference value for the preparation of other positive serum against other poultry virus;Marek′s disease.
