武定鸡和科宝肉鸡成骨细胞矿化及TNF-α基因表达差异研究

摘要 为研究武定鸡和科宝肉鸡成骨细胞分化、矿化及肿瘤坏死因子α(TNF-α)基因表达差异,试验通过提取武定鸡和科宝肉鸡骨间充质干细胞,并诱导分化为成骨细胞,检测成骨细胞分化、矿化差异及TNF-α基因表达差异。结果显示:与科宝肉鸡相比,在骨间充质干细胞诱导为成骨细胞过程中,武定鸡成骨细胞矿化能力较强,且TNF-α基因表达量显著低于科宝肉鸡。研究表明:TNF-α可通过调控成骨细胞分化来影响矿化过程,进而影响骨骼强度。可见,TNF-α基因对家鸡成骨细胞矿化起着重要的作用。 In order to study differences on differentiation,mineralization and TNF-αgene expression of osteoblast in Wuding chicken and Cobb broiler,bone mesenchymal stem cells from Wuding chicken and Cobb broiler were obtained to induce into osteoblasts.Differentiation and mineralization of osteoblast and expression of TNF-αin osteoblasts were detected.The results showed that compared with the Cobb broiler,in the process of induced into osteoblasts,the mineralization ability of Wuding chicken was stronger than that in Cobb broiler.And the expression of TNF-αgene in osteoblasts from Wuding chicken was significantly lower than that in Cobb broiler.It indicated that TNF-αgene could affect bone strength by regulating osteoblast differentiation to influence mineralization process.So,it can be seen that TNF-αgene plays an important role in the mineralization of chicken osteoblasts.
