鸡源铜绿假单胞菌氨基糖苷类耐药特性研究 被引量:3

Detection of Aminoglycoside Antibiotic Resistance and Resistance Genes of Pseudomonas aeruginosa Strains Isolated from Chicken Farms

摘要 为了解铜绿假单胞菌(PA)的氨基糖苷类耐药机制,用纸片扩散法检测分离自鸡场的50株PA对6种氨基糖苷类药物的敏感性,用PCR法检测7种氨基糖苷类修饰酶(AMEs)基因和5种16 S rRNA甲基化酶基因。结果显示:50株PA中45株对氨基糖苷类药物耐药,36株表现出多种氨基糖苷类药物耐药。50株菌中共有45株检测到耐氨基糖苷类药物基因,总检出率为90.0%。AMEs基因aac(3)-Ⅱ、ant(3′′)-Ⅰ、aph(3′′)-Ⅰb、aph(6′)-Ⅰd、aac(6′)-Ⅰb、ant(2′′)-Ⅰ的阳性率分别为88.0%、50.0%、44.0%、24.0%、20.0%、4.0%,16 S rRNA甲基化酶armA基因的阳性率为2.0%,其余5种基因均未检出。提示PA对氨基糖苷类耐药与产AMEs和16 S rRNA甲基化酶有关。 To explore the mechanism of aminoglycoside resistance in Pseudomonas aeruginosa(PA),drug susceptibility test was performed by disk diffusion method(K-B method).7 kinds of aminoglycosides modification enzymes(AMEs)genes and 5 kinds of 16 S rRNA methylase genes among all the isolates were detected by polymerase chain reaction(PCR).Among the 50 strains,forty-five showed drug resistant,and 36 of them showed multiple resistance.Drug resistance genes were detected in 45 strains with a total detection rate of 90.0%.Six kinds of AMEs gene were detected,the positive rate of aac(3)-Ⅱ,ant(3′′)-Ⅰ,aph(3′′)-Ⅰb,aph(6′)-Ⅰd,aac(6′)-Ⅰb,and ant(2′′)-Ⅰwas 88.0%,50.0%,44.0%,24.0%,20.0%,and 4.0%,respectively.The 16 S rRNA methylase gene armA was detected with the positive rate of 2.0%.No other five genes were detected.In this research,the aminoglycoside resistance mechanism of PA was associated with AMEs and 16 S rRNA methylase.
