miR-26基因家族进化分析及其在鸡不同组织中表达研究 被引量:1

摘要 研究旨在探明miR-26基因家族(miR-26s)系统进化历程及其在鸡生殖周期内的表达变化规律。通过文献和miRBase检索脊椎动物的miR-26s序列,利用Ensembl数据库信息,确定了miR-26s在基因组中的位置,采用MEGA 10.0构建了miR-26s的系统进化树;并利用RT-qPCR检测了鸡卵巢组织miR-26s在不同组织及产蛋时期的表达变化。结果显示:在34个脊椎动物中共搜索102条miR-26s序列,miR-26s主要成员(miR-26a、miR-26b、miR-26c、miR-1297和miR-4465)在脊椎动物中分布广泛,且高度保守;均位于基因间隔区(Intergenic region,IGR)。RT-qPCR结果显示,miR-26s在鸡的卵巢基质中表达显著高于其他组织器官,在开产前、开产后和产蛋高峰期卵巢组织中表现出相同规律的表达,即在产蛋期、产蛋高峰期表达水平显著高于开产前。这表明miR-26家族在进化中高度保守,且在调控鸡繁殖调控中发挥了一定的作用。 This experiment was conducted to investigate the miR-26 gene family evolutionary process and its expression regularity in different tissues.The sequences of miR-26 genes were searched in literature and miRBase.They were located by Ensemble database,and their phylogenetic tree was constructed by MEGA 10.0.In addition,the expression levels of miR-26 family were detected in different tissues and stages in chicken by RT-qPCR.A total 102 sequences in 34 species of vertebrate which indicated the extensive existence of miR-26 s(miR-26 a,miR-26 b,miR-26 c,miR-1297 and miR-4465)in different species.The analysis of gene localization showed all miR-26 genes located on the intergenic region.Besides,the similar expression of miR-26 s were detected in covary of pre-laying stage,laying stage and the egg laying peak stage.More miR-26 s mRNAs were detected at laying stage and the egg laying peak stage than that in pre-laying stage.It′s suggested that miR-26 s were highly conservative in evolution process and played a role in regulation reproductive activity in chicken.
