摘要
对中国热带农业科学院油茶研究中心油茶资源圃采集到的油茶叶斑病叶片进行病原菌分离、鉴定,并研究其生物学特性。通过对病原菌培养特性、形态特征的观察及rDNA的转录间隔区(ITS)序列测定,与Genbank中同源性较高的菌株进行序列对比,最后将菌株20-1确定为可可毛色二孢菌(Lasiodiplodia theobromae)。生物学特性研究表明,20-1菌丝在PDA培养基上生长最快,菌丝生长的最适宜温度是20~35℃,pH值及光照对菌丝生长的影响均不大,果糖为最佳碳源,硝酸钠为最佳氮源。
Leaf spot disease survey was taken on pathogen isolation and identification of Camellia oleifera in Camellia Research Center,Chinese Academy of Tropical Agricultural Sciences.The pathogen was identified through culture characteristics,morphological characteristics and genetic molecular analysis.The sequence was compared with the highly homologous strains in Genbank,and the results showed that the pathogen 20-1 was Lasiodiplodia theobromae.In vitro culture showed that PDA medium was the best for the mycelial growth of pathogen.Temperature ranged from 20 to 35℃were optimum for its mycelia growth.The effects of pH and light intensity on the growth of mycelium were not obvious.And fructose was the best carbon source and sodium nitrate was the best nitrogen source.
