青花菜DH系带柄子叶高频再生体系的建立 被引量:1

Establishment of High-frequency Regeneration System from Cotyledons with Petiole in DH Lines of Broccoli

摘要 以青花菜DH系3-6、3-7和3-9带柄子叶为外植体,研究其高频离体再生体系,比较了不同基因型、不同质量浓度植物生长调节剂、不同时期苗龄外植体及不同浓度AgNO3添加剂对不定芽再生频率的影响。结果筛选出3-6和3-7两个基因型:8d苗龄外植体不定芽的再生频率分别为96.9%和97.8%(培养基:MS+0.02mg·L-1NAA+1.0mg·L-16-BA+4mg·L-1AgNO3+10g·L-1琼脂+30g·L-1蔗糖),平均外植体的再生芽数分别为15、14个,生根培养诱导生根率均为100%(培养基:1/2MS+0.05mg·L-1NAA+7g·L-1琼脂+30g·L-1蔗糖),驯化移栽成活率均达100%。 The high frequency of shoot regeneration from cotyledons with petiole in broccoli(Brassica oleracea L. var. italica Plenck)was studied and the effect of the different genotypes,different mass concentration plant growth regulator,different age of explants and different concentration AgNO3 on regeneration frequency of adventitious buds were studied.The results showed that 2 DH lines,3-6 and 3-7,obtained high shoot differentiation rate(96.9% and 97.8%,respectively),after the explants were pre-cultured for 8 days,then placed in the MS medium supplemented with 0.02 mg·L-1 NAA,1.0 mg·L-1 6-BA,4 mg·L-1 AgNO3,10 g·L-1 agar and 30 g·L-1 sucrose.The frequence of shoot regeneration was 15 and 14.The feasible rooting medium was MS + 0.05 mg·L-1 NAA +7 g·L-1 agar + 30 g·L-1 sucrose and the rooting rate reached to 100%.
