摘要
利用SRK基因激酶区的3对特异性引物,对23份甘蓝一代杂种进行PCR扩增,经过克隆测序、Blast分析初步鉴定甘蓝一代杂种的S单元型。结果表明:供试23份材料中共出现了11个S单元型,分别是S7、S16、S28、S35、S45、S57、S68和未知的Sn等8个Ⅰ类S单元型及S2、S5、S15等3个Ⅱ类S单元型。其中,Ⅱ类S单元型的S5出现频率最高,达到19.6%;其次为Ⅰ类S单元型的S28,达到17.4%。相比甘蓝类作物中存在的50多个S单元型而言,本试验材料涉及的S单元型并不多,预示在遗传育种中利用更多的S单元型是十分必要的。
DNA fragments corresponding to kinase domain of S locus kinase(SRK)gene were amplified by PCR with 3 pairs of specific primers in all of the 23 cabbage(Brassica oleracea L. var. capitata L.)F 1 hybrids.The S haplotypes of cabbage F 1 hybrid were preliminary identified via DNA sequencing and Blast analysis.The results indicated that 11 S haplotypes were found in cabbage F 1 hybrid,including class Ⅰ haplotypes,S7,S16,S28,S35,S45,S57,S68 and an unknown one named Sn,and class Ⅱ haplotypes S2,S5 and S15.The highest frequency of S haplotyes was S5 with 19.6% and followed by S28 with 17.4% in this study.Compared over 50 S haplotypes in Brassica,the 11 S haplotypes in cabbage F 1 hybrid suggests that it is very necessary for exploiting more S haplotypes in cabbage breeding programs.
