摘要
为研究马立克病(MD)抗性选育对鸡马立克病病毒(MDV1)感染后外周血淋巴细胞病毒载量的影响,利用MDV1攻毒后第4、7、10、14、21、28和35天共7次采集并分离的全部试验鸡的外周血淋巴细胞为材料,采用real-time FQ-PCR方法对这些材料中meq基因进行绝对定量分析以确定其病毒载量,用来分析比较MDV1在MD抗性鸡与普通鸡以及HVT疫苗免疫鸡与HVT疫苗未免疫鸡体内的增殖情况。结果显示,普通鸡非免疫攻毒组(B组)的MDV1含量于攻毒后第10(P<0.05)、14(P<0.05)、21(P<0.01)、28(P<0.05)、35(P<0.05)天显著高于MD抗性鸡免疫攻毒组(F组),而其他组间均无显著性差异。结果表明,鸡MD抗性选育结合HVT疫苗接种可显著降低鸡体内的病毒载量,从而降低MDV1感染的危害。
Effects of Marek's disease(MD)resistance selection on virus loads in peripheral blood lymphocytes(PBLs)of chickens after the challenge with Marek's disease virus type 1(MDV1)were investigated.Real-time FQ-PCR was used for absolute quantification of the DNA levels of meq gene of PBLs sampled on 7different time points(on day 4,7,10,14,21,28and 35post-infection)from all birds after the challenge of MDV1.The load of MDV1was detected in PBLs to analyze and compare proliferation of MDV1in resistant/common or/and HVT-vaccinated/ HVT-unvaccinated birds.In result,the load of MDV1 was significantly higher in chickens of B line(common line,HVT-unvaccinated and MDV1-challenged)but significantly lower in chickens of F line(MD-resistant line,HVT-vaccinated and MDV1-challenged)on day10(P<0.05),14(P<0.05),21(P<0.01),28(P<0.05)and 35(P<0.05)post-infection.There was no significant difference between the other groups.In conclusion,virus loads in chickens can be reduced significantly by resistance selection in coordination with vaccination,which can reduce the harm of MDV1infection.
