摘要
全基因组扩增(Whole genome amplification,WGA)技术是一种对全部基因组序列进行非选择性扩增的技术,可以用于富集环境样本中的DNA。该技术已广泛用于DNA文库的建立,肿瘤遗传学研究,产前诊断,法医鉴定。近年来多篇文献力证该技术已成功应用在微生物的鉴定及微生物多样性等分子生态学研究领域。本研究简要介绍WGA技术结合微生物分子生态学技术在感染微生态领域中的应用现状,展望其在感染微生态领域中的发展前景,以期能为今后该技术在这一领域的研究提供科学依据。
Whole genome amplification( WGA) is a powerful technique of non-selective amplification of all genomic sequences to allow an increase of limiting DNA from environmental samples. The technique has been widely used in the DNA library establishment,cancer genetics,prenatal diagnosis and forensic identification. Several recent papers suggest that it should be suitable for evaluation of microbial diversity,and genotyping. Here,the state and trend of WGA combined with molecular microbial ecology techniques applied in the field of infection microecology are reviewed.
