摘要
目的探讨辅助性T细胞9(Th9细胞)及其细胞因子白细胞介素9(IL-9)在毛细支气管炎患儿外周血中的表达及在疾病发展过程中的意义。方法选取2016年11月至2018年4月在滨州医学院附属医院儿科住院的80例毛细支气管炎患儿,分为轻度组(29例)、中度组(27例)、重度组(24例);选取同期25名健康体检儿童为健康对照组。采用流式细胞学及酶联免疫吸附试验法检测急性期及恢复期外周血中Th9细胞比例及IL-9表达水平。结果毛细支气管炎重度组、中度组、轻度组患儿急性期Th9细胞及IL-9表达水平均明显高于健康对照组[(0. 66±0. 18)%、(0. 54±0. 11)%、(0. 43±0. 15)%比(0. 28±0. 08)%,(182±19)、(104±14)、(68±10) ng/L比(32±8) ng/L];且重度组和中度组明显高于轻度组,重度组明显高于中度组,差异均有统计学意义(均P <0. 05)。重度组、中度组、轻度组患儿恢复期血清IL-9水平[(35±7)、(32±8)、(33±8) ng/L]均较急性期明显降低,差异均有统计学意义(均P <0. 05); IL-9在恢复期各组患儿和健康对照组间差异均无统计学意义(均P> 0. 05)。结论毛细支气管炎患儿急性期外周血中Th9细胞和IL-9表达明显升高,且疾病程度越严重其表达水平越高,推测Th9细胞及其细胞因子IL-9在毛细支气管炎的发病过程中起重要作用,且与病情严重程度有关。
Objective To analyze the expression and significance of helper T cell 9( Th9 cell) and interleukin-9( IL-9) in peripheral blood of children with bronchiolitis. Methods Eighty children with bronchiolitis admitted to Binzhou Medical University Hospital from November 2016 to April 2018 were divided into mild group( n = 29),moderate group( n = 27) and severe group( n = 24). Twenty-five healthy children were enrolled as control group. Flow cytometry and enzyme linked immunosorbent assay were used to detect Th9 cell and IL-9 in peripheral blood at acute phase and convalescence. Results Th9 cell ratio and IL-9 level in severe group,moderate group and mild group at acute phase were significantly higher than those in control group [( 0. 66 ± 0. 18) %,( 0. 54 ± 0. 11) %,( 0. 43 ± 0. 15) % vs( 0. 28 ± 0. 08) %;( 182 ± 19),( 104 ± 14),( 68 ± 10) ng/L vs( 32 ±8) ng/L];there were also significant differences among the three bronchiolitis groups( all P < 0. 05). During convalescence of bronchiolitis,IL-9 levels in severe group,moderate group and mild group[( 35 ± 7),( 32 ± 8),( 33 ± 8) ng/L] were significantly lower than those at acute phase( all P < 0. 05);there was no significant difference of IL-9 level among the three bronchiolitis groups and control group( P > 0. 05). Conclusions In the acute phase of bronchiolitis,expressions of Th9 cell and its cytokine IL-9 in peripheral blood significantly increase with the severity of disease. Th9 cell and IL-9 may play an important role in the pathogenesis of bronchiolitis.
