小分子RNA Cdc42 GTPase和微小RNA-363-5p在不同级别胶质细胞瘤中的分布规律 被引量:1

摘要 目的探讨Cdc42 GTPase和微小RNA(miR NA)-363-5p在不同级别胶质细胞瘤中的分布规律。方法常规采集胶质细胞瘤患者术后肿瘤样本,多聚甲醛固定24 h后,用蔗糖溶液浸泡至沉底,冰冻切片后应用免疫组织化学方法检测Cdc42 GTPase的表达;其余样品用液氮保存,实时荧光定量-聚合酶链反应检测Cdc42 GTPase和miR NA-363-5p在不同级别胶质细胞瘤中的表达情况。结果实时荧光定量-聚合酶链反应检测结果显示,随着肿瘤级别(肿瘤临床分期Ⅰ、Ⅱ、Ⅲ、Ⅳ期)的增高,miR NA-363-5p表达量逐渐上调[(1.401±0.515)×10^(-6)、(1.665±0.686)×10^(-6)、(5.792±0.000)×10^(-6)、(8.383±2.858)×10^(-6)],而Cdc42GTPase表达量逐渐下调[(2.218±0.694)×10^(-2)、(2.004±0.236)×10^(-2)、(0.285±0.004)×10^(-2)、(0.071±0.007)×10^(-2)],两两比较差异均有统计学意义(均P<0.05)。结论 miR NA-363-5p的靶基因可能是Cdc42GTPase,且随着miR NA-363-5p表达量的上调,Cdc42 GTPase表达量下调,胶质细胞瘤级别增高。 Objective To investigate the distribution of small RNA Cdc42 GTPase and microR NA(miR NA)-363-5p in different grades of glioma.Methods Glioma samples of human were fixed by paraformaldehyde for 24 h and immersed in sucrose solution until it sank to the bottom.Partial tissues were made frozen section to test the expression of Cdc42 GTPase by immunohistochemistry.The rest samples were stored in liquid nitrogen,quantitative real-time polymerase chain reaction was used to detect expressions of Cdc42 GTPase and miR NA-363-5p in different grades of tumor.Results Polymerase chain reaction showed that the expression of miR NA-363-5p gradually increased[(1.401±0.515)×10-6,(1.665±0.686)×10-6,(5.792±0.000)×10-6,(8.383±2.858)×10-6]and the expression of Cdc42 GTPase was down-regulated[(2.218±0.694)×10-2,(2.004±0.236)×10-2,(0.285±0.004)×10-2,(0.071±0.007)×10-2]with the increase of tumor grade;there were significant differences in multiple comparisons(all P<0.05).Conclusion Cdc42 GTPase may be the target gene of miR NA-363-5p and they changed in an opposite direction of trend;up-regulation of miR NA-363-5p and down-regulation of Cdc42 GTPase indicate high grade of glioma.
