荆芥汤中荆芥最佳煎煮时间的实验研究

Experimental study on the best decoction time of Schizonepetain Schizonepeta decoction

目的:通过研究荆芥后下对荆芥汤中胡薄荷酮含量及镇咳、镇痛、免疫功能的影响,探讨荆芥的最佳煎煮时间,为荆芥特殊煎煮方法的临床应用提供理论依据。方法:按照《中国药典》(2015年版)中荆芥含量测定要求测定胡薄荷酮含量;采用致咳、致痛法观察荆芥后下对小鼠镇咳、镇痛作用的影响,通过分析小鼠CD4^+、CD8^+、IL-1、IL-6水平,探讨荆芥后下对小鼠免疫功能的影响。结果:荆芥煎煮7.5 min时,荆芥汤中胡薄荷酮含量最高,与空白组比较其能够明显改善小鼠痛阈值、咳嗽潜伏期及咳嗽次数(P<0.01),提高小鼠CD4^+、CD8^+、IL-1、IL-6水平(P<0.05)。结论:荆芥后下影响荆芥汤中胡薄荷酮含量及治疗效果,荆芥入解表类汤剂宜采用"后下",最佳时间为5～10 min。

OBJECTIVE To investigate the best decoction time of Schizonepeta by studying the effect of Schizonepeta later-decoction on the content of pulegone in Schizonepeta decoction and its antitussive,analgesic and immune functions,provide a theoretical basis for the clinical application of Schizonepeta decoction.METHODS Content of menthol was determined in accordance with the determination of pulegone in Schizonepeta in the 2015 edition of the Chinese Pharmacopoeia.The effects of Schizonepeta on the antitussive and analgesic effects of mice were observed by means of cough and analgesia.By analyzing the levels of CD4+,CD8+,IL-1 and IL-6 in mice,the effects of Schizonepeta on the immune function of mice were investigated.RESULTS The content of puergonone in Schizonepeta decoction was the highest at 7.5 min.Compared with the control group,it significantly improved the mice pain threshold,cough latency and cough frequency(P<0.01),increased levels of CD4+,CD8+,IL-1,and IL-6 in mice(P<0.05).CONCLUSION After the Schizonepeta affects the menthol content and therapeutic effect of Schizonepeta decoction,it is recommended to use"later-decoction"for the treatment of Schizonepeta,and the best time is 5-10 min.
