抑菌试验结合正交试验优选清咽爽喉喷雾剂的提取工艺 被引量:3

Optimization of Qingyan Shuanghou Spray extraction process by orthogonal experiment combined with antibacterial test

摘要 目的:优选清咽爽喉喷雾剂的提取工艺。方法:采用正交试验设计法和药敏纸片琼脂扩散法,以苦参碱含量、抑菌环直径为指标,优选处方最佳醇提水沉工艺。结果:综合抑菌试验和药物含量评分结果发现,在醇提工艺中,各影响因素的顺序依次为乙醇体积分数(P<0.01)>提取次数(P<0.05)>提取时间(P>0.05)。综合各种因素,最佳提取工艺为以85%乙醇为溶剂,液料比10∶1,回流提取2次,每次2 h,20倍水沉24 h,苦参碱的提取率为75.57%(RSD=2.33%,n=3),抑菌环直径(2.5±0.3) cm (n=3)。结论:该工艺稳定可靠,重现性较好,所得提取物具有较好的抑菌效果,可为该药的后期研发提供参考。 OBJECTIVE To optimize the extraction process of Qingyan Shuanghou Spray. METHODS Alcohol extraction-water precipitation method was optimized by orthogonal design and agar diffusion method with the matrine content and inhibitory ring diameter as parameters. RESULTS The results of comprehensive antibacterial test and drug content score showed that ethanol concentration(P<0.01) > extraction times(P< 0.05) > extraction time(P>0.05). Combined with various factors, the optimum extraction process was as follows: 85% ethanol was used as solvent, the ratio of liquid to material was 10∶1, reflux extraction was performed twice, each time for 2 h, and 20 times water precipitation for 24 h. The extraction rate of matrine was 75.57%(RSD=2.33%,n=3), and the diameter of the inhibitory ring was(2.5±0.3) cm(n=3). CONCLUSION The process is stable and reliable with good reproducibility. The obtained extract has superior antibacterial effect and provides reference for further development of the drug.
