基于近红外光谱法对怀牛膝的定性及定量研究 被引量:1

A qualitative and quantitative study of Radix Achyranthes bidentata based on near infrared spectroscopy

摘要 目的:建立怀牛膝近红外(NIR)定量及定性分析模型,实现怀牛膝中齐墩果酸含量的快速测定;快速鉴别不同规格、不同采收地怀牛膝。方法:采集怀牛膝药材样品的近红外光谱。用高效液相色谱(HPLC)法测定样品中齐墩果酸的含量,将所测的齐墩果酸含量与采集的近红外光谱相关联,运用偏最小二乘法(PLS)建立怀牛膝药材中齐墩果酸定量分析模型;运用判别分析法,建立不同采收地、不同规格怀牛膝定性分析模型。结果:定量模型:R^2为0.969 88,RMSEC为0.030 1,RMSEP为0.028 4;定性模型:不同采收地、不同规格怀牛膝定性模型分离均较好,所建模型稳定、可靠。结论:所建立的定性和定量模型准确、可靠,可快速评价不同采收地、不同规格怀牛膝药材的品质。 OBJECTIVE To establish the qualitative and quantitative analysis models of near infrared spectroscopy(NIR),for rapid determination of oleanolic acid contents in Radix Achyranthes bidentata;to identify Radix Achyranthes bidentata with various specifications and harvesting places rapidly.METHODS The NIR spectrum of Radix Achyranthes bidentata samples was collected.With HPLC method,oleanolic acid contents in the samples were determined,correlation of the obtained oleanolic acid contents with the NIR spectroscopy results was examined,and the quantitative analysis model of oleanolic acid contents in Radix Achyranthes bidentata was established with partial least squares(PLS).The qualitative analysis model of Radix Achyranthes bidentata from different harvesting places and with different specifications was established by discriminant analysis.RESULTS Quantitative analysis model:R^2 was 0.969 88,RMSEC,0.030 1;and RMSEP,0.028 4.Qualitative analysis model:Radix Achyranthes bidentata from different origins and with different specifications were well identified with the model,and so the model was stable and reliable.CONCLUSION The established qualitative and quantitative analysis models are accurate and reliable,they can be used to quickly evaluate the quality of Radix Achyranthes bidentata from different habitats and with different specifications.
