摘要
目的:研究茜草对苍耳子诱导的人肝细胞系HepaRG的细胞毒性是否产生某种影响。方法:采用MTT法研究苍耳子提取物的HepaRG细胞毒性,并考察加入茜草提取物后对细胞增殖率的影响;采用ALT和AST测试盒和活性氧(ROS)检测试剂盒,分别研究茜草提取物对苍耳子提取物诱导的HepaRG细胞毒性的生化指标变化。结果:苍耳子提取物对HepaRG细胞有一定的毒性作用,其半数抑制浓度约为60 g·mL^(-1),当加入茜草提取物对细胞增殖率有一定的提高作用;茜草提取物对HepaRG细胞中异常升高的ALT、AST和ROS水平有一定的逆转作用,且存在剂量-效应关系。结论:茜草对苍耳子所致肝细胞毒性有一定的保护作用。
OBJECTIVE To explore whether Rubiae Radix et Rhizoma extract(RE)has some effect on against Xanthium Fructus extract(XE)-induced cytotoxicity in human hepatic cell line HepaRG.METHODS MTT method was used to investigate the cytotoxic effects of XE at various concentrations on HepaRG cells,and the effects of RE on proliferation rate of HepaRG cells were also examined ALT and AST test kit and reactive oxygen species(ROS)test kit were adopted to investigate the changes of the biochemical indicators of ALT,AST and ROS levels for cytotoxicity in HepaRG cells after the addition of RE.RESULTS XE exhibited some cytotoxic effect on HepaRG cells,with IC50 of about 60 g·mL-1.The cell proliferation rate was increased after RE addition.RE showed certain reverse effect on abnormally increased ALT,AST and ROS levels,with dose-effect relationships.CONCUSION RE has certain protective effects on XE-induced cytotoxicity in HepaRG cells.
