百香果乙酸乙酯提取物预防化学性肝损伤和免疫性肝损伤的研究

Study on the preventive effect ofethyl acetate extract of Passion fruit on chemicalliver injury and immunological liver injury

目的:探讨百香果乙酸乙酯提取物对化学性肝损伤和免疫性肝损伤是否具有预防作用。方法:将适应性喂养的小鼠随机分为6组:空白对照组、模型对照组、阳性(联苯双酯)对照组(150 mg·kg-1)、百香果乙酸乙酯提取物低剂量值(15 mg·kg-1)、中剂量值(30 mg·kg-1)、高剂量组(60 mg·kg-1),给药14 d后,除空白对照组外,其余各组腹腔注射1%四氯化碳或尾静脉注射ConA15 mg·kg-1。动物模型构造成功后,小鼠禁食不禁水过夜,小鼠眼眶取血,并且颈椎脱臼处死,解剖小鼠取肝脏,测定血清和肝匀浆中的相关生化指标,并做肝脏的组织病理学检查。结果:百香果乙酸乙酯提取物可以预防肝损伤小鼠血清中的ALT、AST水平的升高,升高肝脏抗氧化系统中抗氧化酶CAT、SOD、GSH-Px的活性及其含量,降低过氧化产物MDA、TG的含量。肝脏组织病理学结果表明:百香果乙酸乙酯提取物对小鼠的肝脏有保护作用。结论:百香果乙酸乙酯提取物对化学性肝损伤以及免疫性肝损伤具有预防作用。

OBJECTIVE To investigate whether ethyl acetate extract of Passion fruit has a preventive effect on chemical liver injury andimmunological liver injury.METHODS Adaptive fed mice were randomly divided into 6 groups:blank control group,model control group,positive(bifendate)control group(150 mg·kg-1),and ethyl acetate extract of Passion fruit low(15 mg·kg-1),medium(30 mg·kg-1)and high dose group(60 mg·kg-1),after administration for 14 days,1%carbon tetrachloride was injected intraperitoneally or ConA 15 mg·kg-1 was injected intravenously into the ventral and caudal vein in all groups except the blank control group.After the animal models were successfully constructed,the mice were fasted overnight,and the orbits of the mice were bled and sacrificed by cervical dislocation.The liver was dissected and the relevant biochemical indicators in serum and liver homogenate were determined.Histopathologicalexamination of the liver was performed.RESULTS Ethyl acetate extract of Passion fruit could prevent the increase of serum ALT and AST levels,increase the activities and contents of antioxidant enzymes CAT,SOD and GSH-Px,and decrease the contents of MDA and TG.Histopathological results of liver showed that the ethyl acetate extract of Passion fruit had protective effects on the liver of mice.CONCLUSION Ethyl acetate extract of Passion fruit has preventive effect on chemical liver injury and immunological liver injury.
