DNA-PKcs抑制剂香兰素对宫颈癌患者细胞放化疗增敏效果探讨 被引量:1

Chemo-and radio-sensitization effects of DNA-PKcs inhibitor vanillin on human cervical cancer cells

摘要 目的探讨DNA依赖蛋白激酶催化亚单位(DNA-PKcs)抑制剂香兰素作用下宫颈癌Hela细胞对顺铂或电离辐射的敏感性,研究DNA-PKcs抑制剂对宫颈癌的放化疗增敏作用。方法采用四氮唑蓝还原法(MTT)检测香兰素联合顺铂对Hela细胞的增殖抑制作用;采用克隆形成试验(平板法)检测香兰素联合电离辐射对Hela细胞的增殖抑制作用。结果香兰素明显增加顺铂对Hela细胞的生长抑制作用,并呈一定剂量依赖性;香兰素也明显增加Hela细胞对电离辐射的敏感性,放射增敏比(SERDq)=1. 57。结论 DNA-PKcs抑制剂香兰素可能对顺铂治疗宫颈癌有增敏作用,也可能对宫颈癌放射治疗有增敏作用。 Objective To observe the sensitivity of cervical cancer cell Hela to cisplatin or ionizing radiation under the action of DNA-dependent protein kinase catalytic subunit(DNA-PKcs)inhibitor vanillin,aiming to investigate the chemo-and radio-sensitization effects of DNA-PKcs inhibitor on cervical cancer cells.Methods The proliferation inhibition effect of vanillin combined with cisplatin on Hela cells was detected by tetrazolium blue reduction method(MTT).The proliferation inhibition effect of vanillin combined with ionizing radiation on Hela cells was investigated by colony formation assay(plate method).Results Vanillin significantly increased the growth inhibitory effect of cisplatin on Hela cells in a dose-dependent manner;vanillin also significantly increased the sensitivity of Hela cells to ionizing radiation with a sensitization ratio(SERDq)of 1.57.Conclusion DNA-PKcs inhibitor vanillin may have chemo-and radio-sensitization effects in the treatment of cervical cancer.
