摘要
目的研究氯胺酮舒张大鼠胸主动脉的作用及机制。方法采用离体血管环灌流装置,观察氯胺酮预对收缩状态下的胸主动脉的作用并探讨其机制。结果氯胺酮(0.01-1mmol·L-1)显著舒张内皮完整的胸主动脉环,而轻微舒张去内皮的胸主动脉环。孵育TEA(10mmol·L-1)、Bacl2(1mmol·L-1)及L-NAME(0.1mmol·L-1)减弱氯胺酮的舒张作用,预孵4-AP(1mmol·L-1)、Gly(0.01mmol·L-1)及Indo(0.01mmol·L-1)对氯胺酮的舒张作用无影响。结论 BKCa、Kir及一氧化氮合酶参与氯胺酮显著舒张胸主动脉的作用,该作用呈浓度和内皮依赖性。
Objective Aim To give an insight into the mechanisms of ketamine induced thoracic aorta rings dilation. Methods Using isolated vascular rings perfusion device to observe the effects of ketamine during precontracted state,and investigate its mechanisms. Results Ketamine( 0.01-1 mmol·L-1) markedly dilated thoracic aorta rings with intact endothelium and slightly dilated thoracic aorta rings with denuded endothelium. Preincubated TEA( 10 mmol·L-1) ,Bacl2( 1mmol·L-1) and L-NAME( 0. 1mmol·L-1 ) decreased the vasodilation effect of ketamine,Preincubated 4-AP( 1mmol·L-1) ,Gly( 0. 01mmol·L-1) and Indo( 0. 01mmol·L-1) had no influences on the vasodilation effect of ketamine. Conclusions BK Ca ,Kir and nitricoxide synthase involved in the effect of ketamine dilated the isolated thoracic aorta rings,and this effect is concentration and endothelium dependent.
