胰腺癌中lncRNA和mRNA表达谱分析及调控网络研究

摘要 本文研究lncRNAs、mRNAs在胰腺癌中的调控机制。通过对胰腺癌表达谱进行分析,基于生物信息学方法,建立lncRNAs-mRNAs网络,通过获得五个功能显著性模块,网络中结点度较大的HUB节点,这些结点与胰腺癌的发生、发展密切相关。对每个模块进行聚类分析,得到度数小却聚集的集合,这些基因也和胰腺癌密切相关。这些模块中m RNA功能注释又与对炎症、免疫、细胞迁移、细胞增殖和凋亡等有关。表明模块中lncRNAs-mRNAs通过参与多种途径参与胰腺癌的发生及发展过程。通过本文的研究得到lnc RNAs与m RNAs协同作用的一些结果,为今后更深入的研究胰腺癌提供指导和借鉴。 In this article, we research the regulation mechanism of lnc RNAs and m RNAs in pancreatic cancer. Application of bioinformatics method, we constructed a lnc RNA-m RNA network based on the pancreatic cancer expression profile,and obtained five functional modules. In the network, the nodes with large degree are closely related to the occurrence and development of pancreatic cancer. Next, we used cluster analysis for each module to get a small set of clusters in which the nodes are closely related to pancreatic cancer. The function annotations of m RNA in these modules are related to inflammation, immunity, cell migration, cell proliferation and apoptosis. It showed that m RNAs and lnc RNAs in the module participated in the occurrence and development of pancreatic cancer by various ways. In our study, we found some synergistic effect of lnc RNAs and m RNAs, which might provide guidance and reference for further study of pancreatic cancer in the future.
