摘要
目的研究拐枣水提液对酒精所致小鼠急性肝损伤的保护作用。方法用50%乙醇给小鼠灌胃建立急性肝损伤模型后,选取40只小鼠,随机分为正常组,模型组,拐枣水提液低、中、高剂量组(0.5mg/mL、1mg/mL、2mg/mL),每组8只,拐枣水提液低、中、高剂量组分别用0.5mg/mL、1mg/mL、2mg/mL拐枣水提液罐胃,1次/天,0.5mL/次,连用7天;正常组和模型组生理盐水灌胃,1次/天,0.5mL/次,连用7天,测定小鼠血清中AST、ALT、MDA,检测肝组织MDA、SOD活性分析小鼠肝损伤程度。结果与模型组相比,拐枣水提液低剂量组对ALT、AST、MDA、SOD无统计学差异(P>0.05);拐枣水提液中剂量组降低ALT含量具有非常显著的统计学意义(P<0.05),降低MDA的产生非常显著的统计学意义(P<0.01),对AST、SOD无统计学差异(P>0.05);拐枣水提液高剂量组对降低ALT、AST含量有非常显著的统计学意义(P<0.01),降低MDA的产生有非常显著的统计学意义(P<0.01),对SOD活性的提升有显著统计学差异(P<0.05)。结论拐枣水提液各剂量组均降低血清ALT、AST含量,提高SOD活性,降低肝组织MDA的产生,尤以拐枣水提液高剂量组(2mg/mL)效果最佳。拐枣水提液对酒精所致小鼠急性肝损伤有保护作用,饮酒者可多食用拐枣及其制品。
Objective To study the protective effect of water extract of H.dulcis on acute liver injury induced by alcohol in mice. Methods Tank with 50% ethanol to mice stomach after acute liver injury model, selection of 40 mice, were randomLy divided into normal group, model group, Low, medium and high dose groups of Water extract of H.dulcis(0.5 mg/mL, 1 mg/mL, 2 mg/mL), each group of eight, Low, medium and high dose groups of Water extract of H.dulcis, respectively with 0.5 mg/mL, 1 mg/mL, 2 mg/mL Water extract of H.dulcis;r tank stomach, 1 time/day, 0.5 mL/time, stay 7 days;Normal group and model group were treated with normal saline tank stomach for 1 time/day,0.5 mL/time for 7 consecutive days, and the serum AST, ALT and MDA in mice were measured, and the liver tissue MDA and SOD activity were measured to analyze the degree of liver injury in mice. Results Compared with the model group, there were no statistically significant differences in ALT, AST, MDA and SOD in the low-dose Water extract of H.dulcis(P>0.05). The reduction of ALT content in the medium-dose Water extract of H.dulcishad was statistically significant(P<0.05), and the reduction of MDA production was statistically significant(P<0.01). There was no significant difference in AST or SOD(P>0.05). The high-dose Water extract of H.dulcishad significant statistical significance in reducing the content of ALT and AST(P<0.01), the production of MDA was significantly reduced(P<0.01), and the SOD activity was significantly improved(P<0.05). Conclusion The content of ALT and AST in the serum was decreased in each dose-extracting group, the activity of SOD was improved, and the generation of MDA in liver tissue was reduced, especially in the high-dose-extracting group(2 mg/mL). Water extract of H.dulcis has protective effect on acute liver injury induced by alcohol in mice. Drinkers can eat more H.dulcis and its products.
