枳椇水提液改善慢性酒精性肝损伤作用研究

目的研究枳椇水提液改善酒精所致小鼠慢性肝损伤的作用。方法每日用56度白酒灌胃,建立小鼠慢性肝损伤模型,剂量10mL/kg,连续3周。选取40只小鼠,随机分为正常组,模型组,枳椇水提液低剂量组、枳椇水提液中剂量组、枳椇水提液高剂量组(1 mg/mL、2 mg/mL、4 mg/mL),每组8只,枳椇低剂量组、枳椇中剂量组、枳椇高剂量组分别用1mg/mL、2mg/mL、4mg/mL枳椇水提液罐胃,1次/d,0.5 mL/次,连用15 d;正常组和模型组生理盐水罐胃,1次/d,0.5 mL/次,连用15 d,测定小鼠血清中AST、ALT、MDA,检测肝组织MDA、SOD活性分析小鼠肝损伤程度以及肝组织脂肪变性程度。结果与模型组相比,枳椇水提液各剂量组均降低血清ALT、AST含量,提高SOD活性,降低肝组织MDA的产生,阻止肝组织脂肪变性,尤以枳椇水提液高剂量组(4mg/mL)效果最明显。结论枳椇水提液高剂量组对慢性酒精性肝损伤具有保护作用,阻止肝组织脂肪变性进程,有利于慢性酒精性肝损伤者肝细胞修复。

Objective To study the effect of Water extract of Hovenia on improving chronic liver injury induced by alcohol in mice.Methods Mice were treated with 56 degree alcohol every day to establish a mouse model of chronic liver injury.The dose was 10 mL/kg for 3 weeks.Selection of 40 mice,were randomly divided into normal group,model group,Low、medium and high dose groups of Water extract of Hovenia(1 mg/mL、2 mg/mL、4 mg/mL),each group of eight,Low、medium and high dose groups of Water extract of Hovenia,respectively with 1 mg/mL,2 mg/mL,4 mg/mL Water extract of Hovenia tank stomach,1 time/day,0.5 mL/time,stay 15 days;Normal group and model group were treated with normal saline tank stomach for 1 time/d,0.5 mL/time,stay 15 days;The levels of AST,ALT and MDA in serum of mice were measured,and the activities of MDA and SOD in liver tissue and nalyze the degree of liver injury and adipose degeneration in mice.Results Compared with the model group,all the dose groups of the hovenia water extract reduced the content of ALT and AST in serum,increased the activity of SOD,decreased the generation of MDA in liver tissues,and prevented the fatty degeneration of liver tissues,especially the group with the highest dose of hovenia water extract(4 mg/mL)had the most effectivet.Conclusion Hovenia dulcis water extract high dose group has protective effect on chronic alcoholic liver injury,prevent the process of fatty degeneration of liver tissue,and is conducive to the repair of liver cells in patients with chronic alcoholic liver injury.
