七叶皂苷钠联合化疗药对HT-29结肠癌细胞系的作用

Basic research of interaction between sodium aescinate and chemotherapy agent on HT-29 colon cell line

摘要 目的分别检测七叶皂苷钠与不同化疗药物在常规培养环境及缺营养、缺氧培养环境下共同作用于结肠癌HT-29细胞系的特点。方法采用MTT法,在体外培养的HT-29细胞系中,分别检测10种化疗药物与七叶皂苷钠同时和序贯使用时,在不同的培养环境中及不同给药顺序下对HT-29细胞的抑制率。结果常规培养状态下,七叶皂苷钠与多种化疗药存在协同作用;共同孵育或先给予化疗药协同作用最强;缺营养状态下,大多数化疗药在共同孵育或者先给予七叶皂苷钠时表现为协同作用;少数化疗药与七叶皂苷钠呈现出拮抗作用。结论七叶皂苷钠与多种化疗药对HT-29细胞有协同作用,该协同作用与两者的给药次序及培养环境相关,其机制需进一步明确。 Objective To detect the interaction of sodium aescinate acted with different chemotherapy agents in normal and ischemic culture environment on HT-29 colon cell line. Methods MTT assay in cultured HT-29 cell lines in vitro were used to detect the role of sodium aescinate combined with 10 kinds of chemotherapy agents in different administrating sequences in different culture environment. Results Sodium aescinate had synergy effect with a variety of chemotherapy agents; under normal culture state, sodium aescinate and chemotherapy agents were co-incubated or firstly given chemotherapy agents had better synergy effect, to that under ischemic state, most chemotherapy agents had synergy effect with sodium aescinate when co-incubated or firstly given sodium aescinate; but a few chemotherapy agents showed antagonism effect with sodium aescinate. Conclusion Sodium aescinate had synergy effect with a variety of chemotherapy agents in HT-29 cells, such synergy effect related to administrating sequences and culture environment, more research were needed to clarify the mechanism.
