川陈皮素对MC3T3-E1细胞成骨分化影响的实验研究 被引量:1

摘要 目的:检测川陈皮素对MC3T3-E1细胞增殖及成骨分化的作用及机制。方法:采用CCK-8法、碱性磷酸酶(ALP)活性检测及实时定量PCR方法评价川陈皮素对MC3T3-E1细胞增殖、成骨分化及相关基因表达的影响。结果:川陈皮素能促进MC3T3-E1的细胞增殖,刺激细胞ALP活性升高,上调BMP-2、COL-I、OCN、Runx 2的基因表达,BMP-2信号阻断剂(Noggin)能抑制成骨分化基因表达。结论:川陈皮素可促进MC3T3-E1成骨分化及成骨基因表达,BMP-2信号通路可能参与了该过程。 Objective:To investigate the effect and mechanism of nobiletin on the proliferation and osteogenic differentiation of MC3 TS-E1 cells.Methods:CCK-8 assay,alkaline phosphatase(ALP)activity assay and real-time quantitative PCR were used to evaluate the effects of nobiletin on the proliferation,osteogenesis differentiation and related gene expression of MC3 T3-E1 cells.Results:Nobiletin at concentrations of 0.1μM,1μM and 10μM concentration-dependently promoted MC3 T3-E1 proliferation,ALP activity and osteogenic gene expression,including BMP-2、COL-I、OCN、Runx2.The expression levels of gene were decreased in varying degrees after treatment with BMP-2 pathway blocker noggin.Conclusion:Nobiletin could promote osteogenic differentiation and osteogenic gene expression in MC3 T3-E1,and BMP-2 signaling pathway might be involved in this process.
