摘要
目的探讨缩醛磷脂对混合细菌感染大鼠阴道炎的改善效果及可能机制。方法选取雌性Wistar大鼠经阴道先后注入金黄色葡萄球菌和大肠埃希菌以构建混合感染性大鼠阴道炎模型,接种5 d后取感染成功的80只模型大鼠随机分为模型组、凝胶基质组和缩醛磷脂高、中、低剂量组,每组16只。缩醛磷脂高、中、低剂量组分别接受10、5、2.5 mg/kg的缩醛磷脂凝胶阴道给药,每天1次,连续1周。比较各组大鼠的治疗效果(炎症分值、炎症改善率和转阴率)、阴道相关指标(阴道pH、阴道体质量指数和阴道分泌物计分)、氧化应激指标[丙二醛(MDA)和超氧化物歧化酶(SOD)]及炎症指标[肿瘤坏死因子-α(TNF-α)和白细胞介素-8(IL-8)]水平,收集治疗期间阴道灌洗液检测阴道细菌的负荷情况。结果与模型组比较,凝胶基质组大鼠治疗效果、阴道相关指标、氧化应激指标、炎症指标及阴道细菌负荷无显著变化;与模型组和凝胶基质组相比,缩醛磷脂高、中、低剂量组大鼠的炎症分值、阴道pH、阴道分泌物计分、MDA、TNF-α和IL-8水平及阴道细菌负荷均降低,而炎症改善率、转阴率和SOD水平均升高(P<0.05),且缩醛磷脂高剂量组指标优于中、低剂量组(P<0.05)。结论缩醛磷脂对混合感染性大鼠阴道炎具有一定的改善效果,可能与其改善氧化应激和炎症反应有关。
OBJECTIVE To explore the effect of plasmalogens on improvement of vaginitis in rats with mixed infection and its possible mechanisms.METHODS The female Wistar rats were injected with Staphylococcus aureus and Escherichia coli via vagina to construct a mixed infection rat vaginitis model.After inoculation for 5 days,80 successfully established model rats were randomly divided into the model group,the gel matrix group,the high-dose plasmalogens group,the medium-dose plasmalogens group and the low-dose plasmalogens group,with 16 rats in each group.The high-dose plasmalogens group,the medium-dose plasmalogens group and the low-dose plasmalogens group respectively received 10,5,and 2.5 mg/kg of the acetal phospholipid gel via vaginal administration once a day for 1 week.The therapeutic effects(inflammation score,inflammation improvement rate and negative conversion rate),vaginal related indicators(vaginal pH,vaginal body mass index and vaginal secretion score),oxidative stress indexes[malondialdehyde(MDA)and superoxide dismutase(SOD)]and inflammatory indicators[tumor necrosis factor-α(TNF-α)and interleukin-8(IL-8)were observed and compared among the five groups of rats.The vaginal lavage fluid specimens were collected during the treatment so as to detect the bacterial load.RESULTS As compared with the model group,there were no significant changes in the therapeutic effect,vaginal related indexes,oxidative stress indexes,inflammatory indexes and vaginal bacterial load in the gel matrix group.The inflammatory score,vaginal pH,vaginal secretion score,levels of MDA,TNF-αand IL-8 and vaginal bacterial load were lower in the high-dose plasmalogens group,the medium-dose plasmalogens group and the low-dose plasmalogens group than in the model group and the gel matrix group,while the improvement rate of inflammation,negative conversion rate and SOD level were significantly higher in the high-dose plasmalogens group,the medium-dose plasmalogens group and the low-dose plasmalogens group than in the model group and the gel matrix group;the above indexes of the high-dose plasmalogens group were significantly better than those of the medium-dose plasmalogens group and the low-dose plasmalogens group(P<0.05).CONCLUSION Plasmalogen can achieve certain effect on improvement of vaginitis in the rats with mixed infection,which may be associated with the improvement of oxidative stress and inflammatory reactions.
