非小细胞肺癌患者HPV感染与RACK1蛋白表达的相关性研究 被引量:6

摘要 目的探讨活化的蛋白激酶C受体(Receptor for activated Ckinase 1,RACK1)在非小细胞肺癌(Non-Small Cell Lung Cancer,NSCLC)患者人乳头瘤病毒(Human papilloma virus,HPV)感染中的表达情况,分析RACK1蛋白表达与NSCLC患者HPV感染的相关性。方法选取2016年4月-2018年3月曲靖市第一人民医院62份NSCLC病理标本为试验组,同期选取36份癌旁组织为对照组。免疫组化法检测RACK1蛋白及HPV L1壳蛋白表达情况,双变量Spearman相关性分析NSCLC患者HPV感染与RACK1蛋白表达的相关性。结果试验组HPV感染率、RACK1蛋白阳性检出率均高于对照组(P<0.05);HPV感染NSCLC患者中RACK1蛋白阳性检出率高于未HPV感染患者(P<0.05);经Spearman相关性分析检验证实,NSCLC患者HPV感染与RACK1蛋白阳性呈正相关(r=0.547,P<0.001)。结论 HPV感染可能是非小细胞肺癌发生发展的病毒学因素,患者一旦合并HPV感染将上调其RACK1蛋白表达,激活下游信号通路,使细胞发生凋亡抵抗,考虑可将RACK1蛋白早期检测作为非小细胞肺癌患者新型标志物。 OBJECTIVE To explore the expression of receptor for activated C-kinase 1(RACK1)in the non-small cell lung cancer(NSCLC)patients with human papillomavirus(HPV)infection and observe the correlation between the expression of RACK1 and the HPV infection.METHODS Totally 62 pathological specimens of NSCLC that were collected from Qujing First People′s Hospital from Apr 2016 to Mar 2018 were assigned as the experimental group,meanwhile,36 para-carcinoma tissues were chosen as the control group.The levels of RACK1 protein and HPV L1 glutelin were detected by means of immunohistochemical assay,and the correlation between the HPV infection and the expression of RACK1 protein was observed via bivariate Spearman correlation analysis.RESULTS The incidence rate of HPV infection and positive rate of RACK1 protein were significantly higher in the experimental group than in the control group(P<0.05).The positive rate of RACK1 protein of the NSCLC patients with HPV infection was significantly higher than that of the patients without HPV infection(P<0.05).The Spearman correlation analysis showed that the HPV infection in the NSCLC patients was positively correlated with the RACK1 protein.CONCLUSION The HPV infection may be a virological factor for the development of NSCLC,the expression of RACK1 protein will be up-regulated when the patients are complicated with HPV infection,which may activate the downstream signaling pathways and make cells resistant to apoptosis.The RACK1 protein could be considered as a novel marker for early examination of the patients with NSCLC.
