共受体信号传导与HIV

摘要 众所周知, CD4分子是HIV-1感染靶细胞的主要受体, 但单纯CD4分子对于病毒进入细胞既不是充分的, 也不是唯一的途径.例如一些高水平表达CD4蛋白的人类细胞, 包括未分化的CD4+单核细胞, 对HIV的感染并不敏感.人-人细胞杂交以及人CD4+细胞与啮齿动物体细胞杂交的研究提示, 除CD4分子外, 细胞膜上的其他某些分子也是HIV进入细胞所必需的.基于此, 1996年Feng等证明, 趋化性细胞因子受体CXCR4可辅助嗜T细胞性HIV-1(X4株)进入宿主细胞.此后, 很快又证明, CCR5可辅助嗜单核-巨噬细胞性HIV-1(R5株)感染靶细胞.这些受体称为HIV-1入侵靶细胞的共受体(coreceptor), 又称辅助受体.由于目前所发现的HIV的共受体绝大多数均是趋化性细胞因子受体, 因此共受体一般多指趋化性细胞因子受体.
