摘要
目的观察芪蚣抗纤方(QF)及拆方对大鼠免疫损伤性肝纤维化的影响。方法 50只SD雄性大鼠随机分为正常对照组、模型组、QF全方干预组、软坚散结药干预组、活血药干预组,猪血清攻击法诱导免疫损伤性肝纤维化大鼠模型,实验过程8周。免疫组化法检测肝组织转化生长因子-β1(transforming growth factor-β1,TGF-β1)相关表达;免疫组织化学染色检测肝纤维化大鼠TGF-β1和Smad2、Smad3阳性表达。结果药物干预后,治疗组TGF-β1含量明显下降,Smad2、Smad3表达减少,全方比拆方效果明显。结论芪蚣抗纤方及拆方对TGF-βl和Smads家族蛋白有显著调节作用,可干预大鼠肝纤维化形成,全方比拆方效果更明显。预防和治疗肝纤维化除了运用常法活血药与软坚药,益气健脾必不可少。
Objective To observe the effect of Qigong Kangxian Fang(QF)and its broken prescriptions on step in immunologic injury of liver fibrosis in rats.Methods Porcine serum attack method was used to induce immunocomplex induced liver fibrosis rat model,50 male SD rats were randomly divided into five groups:normal control group、model control group、QF intervention group、Softening and Resolving Hard Mass herb intervention group.and promoting blood circulation for removing blood stasis intervention group.The whole experiment process was 8weeks.TGF-β1,Smad2 and Smad3of liver tissue were detected with method of immunohistochemistry.Results With Drug intervention,treatment group showed significantly downregulated effect on anti-liver fibrosis factor TGF-β1levels,and Smad2,Smad3 expression was reduced.The Effect of QF is obvious than that of its broken prescriptions.ConclusionQigong Kangxian Fang(QF)and its broken prescriptions on rat liver fibrosis induced by transforming growth factor(TGF-β1)and Smads family proteins have significant moderatin regulation.The intervention for liver fibrosis in rats,can be achieved and QF effect is obvious than that of its broken prescriptions.Therefore,the prevention and treatment of hepatic fibrosis in addition to the use of ordinary law hemorheologic agent and Softening and Resolving Hard Mass herb,is necessary for of replenishing qi to invigorate the spleen.This has certain directive significance to the clinical treatment of liver fibrosis.
