改良三黄汤对感染耐药大肠杆菌鼠生化免疫影响 被引量:4

Biochemical and Immune Influences of Modified Three Yellow Soups on Mice Inoculated Drug-Resistant E.coli

摘要 目的探讨2副改良三黄汤(MTYSⅠ和MTYSⅡ)对多重耐药大肠杆菌人工感染小鼠生化指标及免疫力的影响。方法选取健康的小鼠80只,随机分为健康组、攻毒组、MTYSⅠ组、MTYSⅡ组。自感染试验菌第1d起,试验组每只小鼠每天分别灌服1mg·mL-1中药制剂提取物0.5mL,连续7d,第8d采血测定生化指标及免疫器官指数。结果 MTYSⅠ和MTYSⅡ中药方剂体外对1号菌株的MIC值分别为62.5mg·mL-1、125mg·mL-1;抑菌直径分别为30.9 mm、15.2 mm。只有MTYSⅠ组鼠的肝指数显著增加(P<0.05),其它不明显。肝功能方面:谷丙转氨酶含量两治疗组极显著低于攻毒组(P<0.05);碱性磷酸酶攻毒组在感染后下降较多,治疗组血清中ALP的含量均显著提高(P<0.05);MTYSⅡ治疗组与阴性组差异极显著(P<0.01)。肾功能方面:尿素氮(BUN)、尿酸(UA)攻毒后呈显著(P<0.05)、极显著增高(P<0.01),治疗后恢复。免疫方面:IgG含量MTYSⅠ治疗组显著降低(P<0.05);IgA攻毒后极显著增加,MTYSⅠ治疗组极显著下降,MTYSⅡ治疗组显著下降(P<0.05),但没有恢复正常;IgM攻毒组极显著增高,MTYSⅠ治疗组显著高于健康组(P<0.05)。其它方面,肌酸激酶含量各治疗组显著减少(P<0.05),淀粉酶(AMY)治疗组显著提高(P<0.05)。结论改良三黄汤体外有较强的抑制耐药大肠杆菌作用,能促进机体免疫器官的发育,显著改变生化指标含量,具有保肝利胆作用。综上所述,MTYSⅠ方剂优于MTYSⅡ方剂。 Objective To investigate the biochemical and immune influence of two doses of Modified Three Yellow Soups from patients (MTYS)on mice inoculated the multiple drug-resistant E.coli.Meth-ods 80 healthy mice were randomized into 4 groups (control group,poison group,MTYSⅠ group,and MTYSⅡgroup).The experiment groups were inoculated with the 0.5 ml extractive of 1 mg/mL MTYS for seven days.On 8th day,blood serum biochemical indicators and immune organ indexes were detected from some mice randomly taken from each group.Results MIC of MTYSⅠand MTYSⅡ were respec-tively 62.5 mg/mL and 125 mg/mL;Bacteriostatic diameter were respectively 30.9 mm and 15.2 mm. The liver index of MTYSⅠ increased significantly (P<0.05),other not obvious.About liver function:ALT contents of MTYSⅠand MTYSⅡgroups were significantly lower than that in poison group (P<0.05);ALP content of poison group was greatly decreased,but ALP in serum of treatment groups was significantly increased (P<0.05),ALP content of MTYSⅡ were extremely different from control group (P<0.01).About renal function:BUN and UA contents of poison group were significantly increased (P<0.05 and P<0.01,respectively),then recovered to normal value after treatment.About immuniza-tion:IgG content of MTYSⅠwas significantly reduced (P<0.05);IgA content of poison group was sig-nificantly increased (P<0.05),IgA contents of MTYSⅠ and MTYSⅡgroups were significantly reduced <br> (P<0.05 and P<0.01,respectively),but not back to normal;IgM content of poison group was signifi-cantly increased (P<0.05),IgM content of MTYSⅠhigher than control group (P<0.05).CK con-tents of all groups were significantly lowered (P<0.05),and AMY content was significantly higher than treatment group (P<0.05).Conclusion MTYS could inhibit drug-resistant E.coli in vitro,promote development of immune organs,significantly change biochemical indicators content ,and protect liver and gallbladder.And MTYSⅠ could have better effect than MTYSⅡ.
