“通督启神”法两种电针对AD模型小鼠行为学及大脑皮层COX-2、TGF-β1表达的影响 被引量:13

摘要 目的:观察"通督启神"法两种电针干预对AD模型小鼠行为学及大脑皮层炎性因子表达的影响,阐释两种电针治疗AD的机理及疗效差异。方法:40只AD模型小鼠随机分为模型组(AD)、药物组(M)、脉冲电针组(EA)、音乐电针组(MEA),同背景C57BL/6小鼠为正常对照组(N),每组10只; EA组、MEA组束缚后针刺印堂、百会20 min,水沟点刺不留针,M组灌胃给药(盐酸多奈哌齐0. 92 mg/kg)后同等条件束缚,AD组和N组同等条件束缚; Morris水迷宫观察小鼠行为学变化,Western blot检测小鼠大脑皮层炎性因子表达情况。结果:EA组、MEA组的逃避潜伏期较AD组均下降(P <0. 01),目标象限游泳时间/总时间、穿越平台次数均增加(P <0. 05或P <0. 01); EA组、MEA组大脑皮层COX-2、TGF-β1的表达较AD组均下降(P <0. 01)。结论:"通督启神"法两种电针干预均能改善AD模型小鼠的空间学习、记忆能力,其机理可能与抑制MG活化、降低大脑皮层COX-2、TGF-β1的表达有关,但两者比较无统计学差异(P> 0. 05)。 Objective: To observe the effect of two kinds of electro-acupuncture on behavior and expressions of inflammatory factors in cerebral cortex of the mice with Alzheimer’s disease( AD) under the guidance of Tongdu Qishen theory,and to explore the mechanism and the difference of therapeutic effect of the two kinds of electroacupuncture on AD. Methods: 40 AD model mice were randomly divided into the model group,the medication group,the pulse current electro-acupuncture group and the music electro-acupuncture group;C57BL/6 mice from the same background were taken as the normal control;10 mice in each group. The mice were bounded,the two electro-acupuncture groups were treated with needling Yintang( EX-HN3) and Baihui( GV20) for 20 minutes,and treated with needling Shuigou( GV26) without retaining the needle. The medication group was treated with Donepezil Hydrochloride( 0. 92mg/kg). Besides the electro-acupuncture groups,the other three groups were given the same conditions of binding. Morris water maze was used to observe the behavioral changes of mice,and Western blot method was used to detect the expressions of inflammatory factors of the cerebral cortex in mice. Results: The escape latency of the two electro-acupuncture groups was lower than that of the model group( P < 0. 01);the target quadrant swimming time/total time and the number of platform crossing in the two electro-acupuncture groups were increased than those of the model group( P < 0. 05 or P < 0. 01). The expressions of COX-2 and TGF-β1 of the cerebral cortex in the two electro-acupuncture groups were lower than those of the model group( P < 0. 01). Conclusion: Under the guidance of Tongdu Qishen theory,the two kinds of electro-acupuncture can improve the spatial learning and memory ability of AD model mice,the mechanism may be related to inhibiting MG activation and decreasing the expressions of COX-2 and TGF-β1in cerebral cortex. There was no significant difference between the two kinds of electro-acupuncture( P >0. 05).
