摘要
目的:探索组胺H3受体激动剂IMETIT对变应性鼻炎(allergic rhinitis,AR)模型豚鼠嗜酸性粒细胞浸润及嗜酸性粒细胞趋化因子含量的影响。方法:30只豚鼠随机均分为3组,分别为对照组、致敏组、药物处理组。对照组以生理盐水,另2组以卵清蛋白为致敏原,复制AR模型,3组均以1%卵清蛋白生理盐水溶液滴双鼻激发,记录30 min内喷嚏、挠鼻次数。AR模型建立后,对照组、致敏组、药物处理组分别以生理盐水(10 m L/kg)、生理盐水(10 m L/kg)、生理盐水(10 m L/kg)+IMETIT(30 mg/kg)灌胃,每天1次,连续5 d,处死。取鼻中隔黏膜组织行HE染色,观察嗜酸性粒细胞浸润程度;免疫组织化学法检测嗜酸性粒细胞趋化因子含量。结果:致敏组和药物处理组30 min内喷嚏次数分别为20.90±4.88、23.10±4.59,30 min内挠鼻次数分别为26.80±4.80、25.00±3.80,均明显高于对照组(P均=0.000)。致敏组、药物处理组每高倍视野下嗜酸性粒细胞浸润数分别为21.60±4.23、22.37±3.08,嗜酸性粒细胞趋化因子阳性表达率分别为(30.16±3.09)%、(29.60±2.88)%,两组间差异均无统计学意义(P分别为0.581和0.676)。结论:IMETIT不能减少AR模型豚鼠鼻中隔黏膜嗜酸性粒细胞浸润数及嗜酸性粒细胞趋化因子含量。
Objective:To explore the influence of histamine receptor 3 (H3 R)agonist,IMETIT,on eo-sinophils and one of eosinophilic chemokines (eotaxin)in allergic rhinitis(AR)model using guinea pigs. Methods:Thirty guinea pigs were randomly divided into three groups:control group,sensitized group with-out treatment and sensitized group with drug treatment.The sensitized group with or without drug treatment were immunized by ovalbumin (OVA)to establish AR model and the control group by saline to contrast, then they were stimulated intranasal by ovalbumin (1 %)respectively.The times of sneezing and scratching nose in 30 min were recorded.The control group and the sensitized group without treatment were given gas-tric gavage by saline.And the sensitized group with drug treatment were given gastric gavage by saline (1 0 mL/kg)with IMETIT (30 mg/kg).Then we investigated the eosinophil infiltration degrees and eosinophilc chemokines content in nasal septum mucosa using HE staining and immunohistochemistry technology.Re-sults:There was significant difference between the control group and the sensitized group with or without drug treatment in the numbers of sneezing and the numbers of scratching nose(P =0.000).There was no significant difference between the sensitized group with or without drug treatment in the means of eosinophil and eotaxin positive expression rate in nasal septum mucosa(P =0.581 ,0.676).Conclusion:The eosin-ophil infiltration and the level of eotaxin of nasal septum mucous could not be reduced by histamine H3 re-ceptor agonist,IMETIT.
