摘要
目的:探讨Polo样激酶1(Polo-like kinase 1,PLK1)在宫颈鳞状细胞癌(squamous cell carcinoma,SQCC)及其癌前病变中的表达及临床意义。方法:选择34例正常宫颈黏膜,82例宫颈SQCC,166例宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)标本,其中CINⅠ级48例,CINⅡ级42例,CINⅢ级76例,采用免疫组织化学法检测PLK1的表达,分析其表达与宫颈SQCC临床病理因素的关系。结果:PLK1在正常宫颈黏膜、CINⅠ级、CINⅡ级、CINⅢ级/原位癌和宫颈SQCC的阳性表达率分别为14.71%、22.92%、35.71%、69.74%和86.59%,差异有统计学意义(χ2=87.42,P<0.01)。PLK1阳性表达率与宫颈癌病理分级和临床分期相关(P均<0.05),而与年龄、高危型HPV感染、盆腔淋巴结转移以及复发和转移无关(P均>0.05)。生存分析显示,PLK1高表达患者总体生存率明显低于PLK阴性和低表达者(P均<0.05)。结论:PLK1可能参与宫颈SQCC的发生发展过程,是判断宫颈SQCC预后的潜在分子标志物。
Objective: To investigate the expression of Polo-like kinase 1( PLK1) in cervical squamous cell carcinoma( SQCC) and its clinical significance. Methods: The immunohistochemical staining method was conducted to detect the expression of PLK1 in 34 cases of cervical squamous epithelium,82 cases of cervical SQCC,166 cases of cervical intraepithelial neoplasia( CIN),including 48 cases of CIN Ⅰ,42 cases of CIN Ⅱ and 76 cases of CIN Ⅲ / carcinoma in situ. And the correlation of PLK1 expression and clinicopathologic factors of cervical SQCC was analyzed. Results: There was significant difference in the expression rate of PLK1 among cervical squamous epithelium,CIN Ⅰ,CIN Ⅱ,CIN Ⅲ / carcinoma in situ and cervical SQCC,which was 14. 71%,22. 92%,35. 71%,69. 74% and 86. 59%,respectively( χ2= 87. 42,P < 0. 01). The overexpression of PLK1 in cervical SQCC was significant correlated with the pathologic grade,FIGO clinical stage( both P < 0. 05),whereas uncorrelated with patient age,high-risk human papillomavirus infection,pelvic lymphatic metastasis and recurrence / metastasis( both P > 0. 05). In cervical SQCC,there was significant difference in the survival curves between the patients with high PLK1 expression and those with low or negative PLK1 expression( P < 0. 05). Conclusion: The PLK1 over-expression might involve in the development,invasion and metastasis and could be used as a prognostic factor of cervical SQCC.
