摘要 目的 :研究原发卵巢上皮癌 (POEC)中乳腺癌转移抑制基因 (BRMS1)和运动相关蛋白 (fascin)基因表达水平及其意义。方法 :用半定量逆转录聚合酶链反应 (RT PCR)技术检测 2 5份正常卵巢、15份良性卵巢上皮瘤、2 8份卵巢上皮癌及 15份卵巢癌盆、腹腔种植转移灶中BRMS1和fascin基因mRNA表达情况。结果 :(1)BRMS1基因表达 :在POEC为 0 .4 73± 0 .179,明显低于正常卵巢和良性上皮瘤组 (P <0 .0 5 ) ;在III、IV期 (晚期 )卵巢癌原发灶中的表达显著低于I、II期 (早期 ) (P =0 .0 12 ) ,在腹腔脱落细胞阳性患者卵巢癌组织中的表达明显低于阴性者 (P =0 .0 4 3) ;(2 )fascin基因表达 :在POEC为 0 .94 1±0 .2 87,明显高于良性上皮瘤的 0 .382± 0 .0 4 6(P <0 .0 0 1) ;其表达与分期有关 ,在III、IV期卵巢癌原发灶中的表达显著高于I、II期 (P =0 .0 2 ) ;(3)fascin/BRMS1的相对光密度值(ROD)比值随肿瘤转移增加。结论 :BRMS1和fascin基因表达与卵巢上皮癌转移密切相关 。 Objective:To investigate the expressions of BRMS1(breast-cancer metastasis suppressor 1) and fascin(an actin-bundling protein) mRNA in primary ovarian epithelial carcinoma (POEC) and their significance.Methods:The expressions of BRMS1 and fascin mRNA in 25 normal ovarian tissues,15 benign ovarian tumors,28 POEC and 15 seeding deposits in peritoneum were assessed by reverse transcription-polymerase chain reaction (RT-PCR).Results:(1)The expressions of BRMS1 in POEC was 0.473±0.179.The expression was lower significantly in POEC than that in normal ovary and benign tumors(P<0.05).The expression of BRMS1 was lower significantly in stage III and IV than that in stage I and II(P=0.012),whereas the expression was lower significantly in POEC with positive shed cells in the abdominal cavity than that in POEC with negative shed cells (P=0.043).(2)The expression of fascin was related to clinical stage.The expression of fascin was lower significantly in earlier stage (I and II) than that in advanced stage (III and IV ) (P=0.02).(3)The relative optic density ratio of fascin to BRMS1 increased with the development of metastasis.Conclusions:Expressions of BRMS1 and fascin in POEC are associated with clinical metastasis of ovarian carcinoma.The increasing of fascin/BRMS1 could be an index for metastases in POEC.
