与肺癌呈负相关表达的cDNA序列鉴定

摘要 目的 :通过进一步鉴定mRNA差异显示获得肺癌差异表达的cDNA序列 ,为克隆肺癌相关基因打下基础。方法 :采用逆转录酶聚合酶链反应 (RT PCR)、Northern印迹杂交和Southern印迹杂交方法检测表达序列标签 (EST)LXDD1在肺癌组织和肺癌细胞系中的表达及缺失。 结果 :RT PCR、Northern印迹杂交证实LXDD1序列在肺癌及正常肺支气管粘膜组织中表达存在差异 ,Southern 印迹杂交结果显示其在肺癌组织中未见缺失。 结论 :LXDD1在肺癌组织中表达下调 ,提示其缺失可能与肺癌的发生。 Objectives: To further identify differentially expressed cDNA sequences in human lung carcinoma isolated by mRNA differential display method for cloning lung carcinoma related genes. Methods: Reverse transcription polymerase chain reaction (RT PCR),Northern blot and Southern blot were employed to detect the expression and deletion of Expressed Sequence Tag(EST) LXDD1 in human lung carcinoma and lung cancer cell lines. Results: Further RT PCR analysis and Northern blot showed that LXDD1 was expressed much lower in lung carcinoma tissue than that in adjacent normal tissues of trachea epithelia. Southern blot showed no deletion of EST (LXDD1) in lung carcinoma. Conclusions: These results suggested that EST (LXDD1) might be involved in the pathogenesis of lung carcinoma.
