敲低hnRNP H基因对子宫内膜癌Ishikawa细胞增殖和细胞周期的影响及意义 被引量:1

摘要 目的探讨不均一核糖体蛋白H(hnRNP H)siRNA敲低hnRNP H基因表达及其对子宫内膜癌Ishikawa细胞增殖和周期的影响。方法将hnRNP H基因的小干扰RNA(siRNA)转染至子宫内膜癌Ishikawa细胞株,细胞分对照组、阴性对照、siRNA组,通过实时定量PCR和Western印迹检测hnRNP H mRNA和蛋白水平,以确定siRNA转染效率。通过MTT实验检测各组细胞增殖情况;Western印迹检测三组细胞增殖蛋白PCNA核增殖抗原和细胞周期cyclin D1、CDK4的蛋白水平。结果敲低子宫内膜癌Ishikawa细胞hnRNP H基因的表达能有效抑制肿瘤细胞的生长,使增殖蛋白PCNA下降,下调细胞周期蛋白cyclin-D1、CDK4的表达。结论敲低hnRNP H基因可能是子宫内膜癌基因治疗靶点。
