齐墩果酸通过线粒体途径诱导原髓细胞白血病细胞株细胞凋亡 被引量:3

The effect of Oleanolic acid to HL-60 cell through mitochondria pathway

摘要 目的探讨原髓细胞白血病细胞株HL-60细胞半胱氨酸蛋白酶(Caspase)-9、Smac/DIABLO、细胞色素C(Cyt-c)和线粒体凋亡诱导蛋白(Bid)表达及齐墩果酸(OA)对HL-60细胞线粒体凋亡信号通路的干预作用。方法细胞分瓶培养,实验设空白对照组和12、24、48 h的OA用药组,收集细胞,提取细胞总蛋白、线粒体蛋白和去线粒体细胞浆蛋白,Western印迹法分别检测Caspase-9、Bid、Smac/DIABLO和Cyt-c的表达。结果OA引起HL-60细胞质中Cyt-c和Smac/DIABLO含量增加,并引起Caspase-9的激活,线粒体Bid蛋白含量增加。结论 OA可以通过线粒体凋亡信号通路诱导HL-60细胞凋亡。 Objective To study the effect of Oleanolic acid to HL-60 cell through mitochondria pathway. Methods The cells were divided into blank control and OA treatment groups,extracted total protein,the expression of Caspase-9 was assayed by Western blot. HL-60 cells were collected from blank control and OA treatment groups,extracted mitochondria from cytoplasm,the expressions of Smac / DIABLO,Cyt-c,Bid were assayed by Western blot. Results OA increased the expressions of Cyt-c and Smac / DIABLO in HL-60 cells,and activated Caspase-9,induced apoptosis by mitochondrial pathway. Bid protein increased in the mitochondria,participate in OA-induced apoptosis in HL-60 cells. Conclusions OA induce apoptosis of HL-60 cells through mitochondrial pathway.
