Aβ_(25~35)对大鼠海马神经元及小胶质细胞的影响 被引量:2

摘要 目的探讨双侧海马注射Aβ25~35不同时间点对大鼠海马神经元及MG的影响。方法雄性Wistar大鼠随机分为对照组、模型组(注射Aβ后2、7、15、21 d 4个时间点),双侧海马注射Aβ建立大鼠AD模型。水迷宫法观察大鼠学习记忆能力;硫堇染色观察海马CA1区神经元和胶质细胞;Aβ、CD11b免疫组化染色分别观察Aβ沉积和活化MG。结果与对照组相比,模型组大鼠在第3~5天逃避潜伏期均显著延长,学习记忆能力下降(P<0.05);4个时间点模型组大鼠海马都有神经元缺失,胶质细胞增生,2 d、7 d、15 d组大鼠神经元损伤呈加重趋势,但21 d组神经元损伤较15 d组有减轻趋势(P<0.05);4个时间点模型组大鼠都有Aβ沉积和活化的MG,Aβ沉积量及MG活化自第7天后呈下降趋势(P<0.05)。结论 Aβ可以引起神经元损伤,降低大鼠的学习记忆能力;Aβ可以激活MG,引起MG形态和数量改变,MG可吞噬、清除Aβ;Aβ沉积量和MG活化数量变化呈正相关。
