烟酰胺单核苷酸腺苷酰转移酶1基因对体外原代培养神经元轴突发育的影响

摘要 目的评价烟酰胺单核苷酸腺苷酰转移酶(NMNAT)1基因对体外原代培养小鼠神经元轴突发育的影响。方法在体外建立了原代培养小鼠神经元模型,并通过RNA干扰和基因过表达的方法控制NMNAT1基因的表达水平。应用电穿孔转染,上调或下调NMNAT1基因的表达水平,应用免疫细胞化学方法研究神经元轴突形态。在所有的功能试验中,把一种高效的特定的非竞争性NMNAT1抑制剂FK-866(CAS658084-64-1)用作药物学对照。结果在体外原代培养的皮层神经元中下调NMNAT1基因能减缓轴突生长,同时引起轴突分支的减少。结论 NMNAT1基因在原代培养的皮层神经元形态发生中起显著作用,表明NMNAT1基因的丢失可能会引起中枢神经系统神经元变性。
