摘要
目的观察姜黄素对大鼠子宫内膜异位症(EMS)雌激素生成代谢酶的影响。方法建立SD大鼠子宫内膜异位症动物模型,3 w后,将其随机分为姜黄素高剂量组(n=9)、中剂量组(n=8)、低剂量组(n=9)、孕三烯酮组(n=9)、模型组(n=10)及假手术组(n=10)。给药3 w后,发光免疫分析法(ECLIA)检测异位组织雌二醇(E2);ELISA检测组织前列腺素E2(PGE2)、血清与腹腔液白介素-1β(IL-1β);免疫组化(SP法)测定组织芳香化酶P450(P450arom)、环氧化酶(COX)-2蛋白表达。结果与模型组比较,姜黄素各剂量组上述指标含量均显著下降(P<0.05)。结论姜黄素通过下调大鼠局部病灶雌激素生成代谢酶,改善体内高雌激素状态而治疗EMS。
Objective To explore the effect of Curcumin on the metabolic enzymes of estrogen and related inflammatory cytokine in endometriosis( EMS) rat models. Methods EMS model was established in SD rats. After 3 weeks,they were randomized divided into high( n=9,100 mg/kg),middle( n = 8,50 mg/kg),low dose of Curcumin( n = 9,25 mg/kg),gestrinone( n = 9,0. 5 mg/kg),model( n = 10,20 mg / kg) and sham groups( n = 10,20 mg / kg). After 3 weeks of treatment,the level of E2 in ectopic endometrial tissue was measured by ECLIA; the content of PGE2 in ectopic endometria,IL-1β in serum and peritoneal fluid were determined by ELISA. The expressions of protein of P450 arom and COX-2 in ectopic endometria were determined by immunohistochemistry. Results Compared with model group,above indexes of every dose of Curcumin groups were decreased significantly( P<0. 05). Conclusions Curcumin could improve the environment of high level of estrogen and abdominal cavity to treat EMS by inhibiting the positive feedback loop of estrogen and IL-1β in local lesions.
