丝胶对2型糖尿病大鼠肝脏肝细胞核因子-4α表达的影响 被引量:1

摘要 目的研究丝胶对2型糖尿病大鼠肝脏肝细胞核因子(HNF)-4α表达的影响。方法雄性SD大鼠48只随机分为:正常对照组、糖尿病模型组、丝胶治疗组和丝胶预防组,每组12只大鼠。链脲佐菌素连续腹腔注射制作2型糖尿病大鼠模型,待模型成功建立后,模型组大鼠不做任何处理,丝胶治疗组大鼠给予2.4 g·kg-1·d-1的丝胶连续灌胃35 d;丝胶预防组大鼠在链脲佐菌素连续腹腔注射前,给予丝胶(2.4 g·kg-1·d-1)灌胃,时间与丝胶治疗组相同。分别采用Western印迹法和RT-PCR法检测大鼠肝脏HNF-4α蛋白和mRNA的表达。结果与正常对照组大鼠比较,糖尿病模型组大鼠肝脏HNF-4α蛋白和mRNA的表达明显升高(P<0.01);与糖尿病模型组大鼠比较,丝胶治疗组、丝胶预防组大鼠肝脏HNF-4α蛋白和mRNA的表达明显降低(P<0.01)。结论丝胶改善糖代谢的作用可能与其调节肝脏HNF-4α的表达有关。 Objective To study the effects of sericin on hepatocyte nuclear factor 4 alpha( HNF-4α) expression in liver of type 2diabetes mellitus rats model. Methods 48 male SD rats were randomly divided into the following 4 groups: normal control,diabetes model,sericin treatment and sericin prevention groups( n = 12). The type 2 diabetes model rats were induced by continuous intraperitoneal injection of streptozotocin. After successfully establishing the diabetes animal models,the rats in diabetes model group were not given any more treatments. Rats in sericin treatment group were lavaged with sericin( 2. 4 g·kg-1·d-1). Rats in sericin prevention group were lavaged with sericin( 2. 4 g·kg-1·d-1) before injecting streptozotocin. Western blot and RT-PCR were used to detect the HNF-4α protein and mRNA expression in liver. Results The HNF-4α protein and mRNA expression in liver of rats in model group were obviously higher than those of normal control group( P<0. 01). The HNF-4α protein and mRNA expression in liver of rats in sericin treatment group and sericin prevention group were obviously lower than those of model group( P<0. 01). Conclusions The effects of sericin on improving glucose metabolism may be associated with its down-regulating the HNF-4α expression in liver.
