慢性阻塞性肺病大鼠血红素加氧酶-1表达及其临床意义 被引量:1

摘要 目的探讨血红素加氧酶(HO)-1在慢性阻塞性肺病(COPD)大鼠肺组织中的表达及其临床意义。方法 Wistar健康雄性大鼠60只随机分为模型组、药物干预组、生理盐水对照组各10只,分别给予烟熏配合脂多糖造模,烟熏配合脂多糖造模+大蒜素6.17 mg/kg灌胃,正常喂养等干预措施,5 w后进行肺功能测定,包括动脉血氧分压(Pa O2)、二氧化碳分压(Pa CO2)、氧合指数(Pa O2/Fi O2)。同时光镜下观察肺脏的形态,采用RT-PCR法检测各组HO-1 m RNA的表达。结果与生理盐水对照组比较,模型组3 s用力呼气容积(FEV)3.0/用力肺活量(FVC)、呼气峰流速(PEF)、Pa O2、Pa O2/Fi O2、HO-1蛋白及基因表达下降显著(P<0.01),Pa CO2升高显著(P<0.01)。与模型组比较,药物干预组FEV3.0/FVC、PEF、Pa O2、Pa O2/Fi O2、HO-1蛋白及基因表达升高显著(P<0.01),Pa CO2下降显著(P<0.01)。各组光镜下肝脏显示:生理盐水对照组肺结构清晰,肺泡间毛细血管轻度扩张,部分支气管旁间质存在少许淋巴细胞浸润;模型组肺组织结构模糊,肺泡间毛细血管扩张明显并伴有出血,见大小不一凝固性肺坏死灶;药物干预组肺组织结构尚清,少量毛细血管扩张充血,肺泡腔内存在少许水肿液。结论 HO-1可能在COPD的发病过程中起保护作用,大蒜素改善COPD大鼠肺功能结构。
